Role of Methionine Sulfoxide Reductases A and B of Enterococcus faecalis in Oxidative Stress and Virulence

Abstract: Methionine sulfoxide reductases A and B are antioxidant repair enzymes that reduce the S-and R-diastereomers of methionine sulfoxides back to methionine, respectively. Enterococcus faecalis, an important nosocomial pathogen, has one msrA gene and one msrB gene situated in different parts of the chromosome. Promoters have been mapped and mutants have been constructed in two E. faecalis strains (strains JH2-2 and V583) and characterized. For both backgrounds, the mutants are more sensitive than the wild-type par… Show more

“…The E. faecalis V19 parental strain (Table 1) used in this study is a plasmid-cured derivative of the vancomycin-resistant clinical isolate V583 (38). E. faecalis V19 and its derivates were cultivated at 37°C in M17 medium (36) supplemented with 0.5% glucose (GM17) or in M17 MOPS (morpholinepropanesulfonic acid) medium supplemented with glycerol as the sole carbohydrate (17).…”

“…The parental E. faecalis strain used in this work was strain V19, a plasmid-cured derivative of the vancomycin-resistant clinical isolate V583 (38). E. coli XLIBlue was used as the recipient for cloning.…”

“…All three are involved in resistance to H 2 O 2 , to various degrees, while survival in murine macrophages was mostly affected in the ⌬tpx mutant and the peroxidase triple mutant. Two additional antioxidant repair enzymes, the methionine sulfoxide reductases MsrA and MsrB, have been characterized for E. faecalis (526). Mutants carrying a deletion in the respective genes exhibited sensitivity to H 2 O 2 and attenuated virulence in systemic and urinary tract infection models.…”
