Role of neutrophils, platelets, and extracellular vesicles and their interactions in COVID‐19‐associated thrombopathy

Abstract: The COVID‐19 pandemic extended all around the world causing millions of deaths. In addition to acute respiratory distress syndrome, many patients with severe COVID‐19 develop thromboembolic complications associated to multiorgan failure and death. Here, we review evidence for the contribution of neutrophils, platelets, and extracellular vesicles (EVs) to the thromboinflammatory process in COVID‐19. We discuss how the immune system, influenced by pro‐inflammatory molecules, EVs, and neutrophil extracellular tra… Show more

“…Interactions between platelets and neutrophils are crucial for inflammation and immunity [ 61 ]. Currently, the platelet–neutrophil crosstalk is well characterized ( Figure 1 ), both in physiologic and pathologic conditions, where the thromboinflammatory response is often described as “a vicious cycle” [ 62 ]. Upon vessel wall damage, platelets are activated, adhere, change their shape, secrete their granule contents and start to form aggregates [ 63 ].…”

“…Accumulated clues to the puzzle of SARS-CoV-2 have suggested that when a virus can initiate platelet and neutrophil driven exacerbated macrophage inflammatory circuits, this can spell a pandemic. The molecules of the purinergic signalling pathway receptosome are directly implicated as key mediators for the interactions between cells involved in these principle pathological processes of concern (223)(224)(225)(226). Despite available vaccines and emergency use antibodies (EUA), a pharmacological prevention and treatment therapy of COVID-19 is still urgently needed, especially for PACS sequelae.…”

“…TF-harboring cells shed high amounts of EVs from their surfaces. EVs, in turn, are associated with increased plasmatic thrombin generation and thromboembolic events, with endothelial cytokine release and leukocyte activation in COVID-19 patients [ 153 ].…”
