Role of Nitric Oxide in Chronic Allergen-Induced Airway Cell Proliferation and Inflammation

Abstract: Chronic cellular inflammation and airway wall remodeling with subepithelial fibrosis and airway smooth muscle thickening are features of chronic asthma. We determined the role of nitric oxide in the pathogenesis of allergen-induced airway cell proliferation and inflammation by studying the effects of a relatively selective prodrug inhibitor of nitric-oxide synthase type 2 (NOS2), L-N 6 -(1-iminoethyl)-lysine-5-tetrazole amide (SC-51). Brown-Norway rats were sensitized to ovalbumin and were exposed to ovalbumin… Show more

“…While the function of iNOS in the process of airway inflammation has been addressed in a number of reports [31][32][33][34], its exact role remains elusive. This is due primarily to the different roles that its product (NO) plays during inflammation.…”

“…Also, the role of nitric oxide, a previously characterized mediator of allergic airway inflammation (45,46) was studied in a guinea-pig of allergic conjunctivitis and a series of experiments demonstrated a major role of this gaseous molecule in regulating the vascular permeability and stimulating prostaglandin E2 production (47,48). As with other species, some guinea-pig strains may be more subtle to allergen sensitization and challenge.…”

“…All major laboratory rat strains have been used to establish allergic conjunctivitis models. Next to Brown-Norway rats (60,61), which are also the preferred strain for the induction of allergic airway inflammation (45,46) IgE-mediated models. IgE-mediated models of rat conjunctivitis used either direct or indirect sensitization and a large number of questions were answered with this species.…”

“…These interactions are orchestrated by a large number of neuromediators such as tachykinins (6), CGRP, vasoactive intestinal polypeptide (VIP), gaseous molecules such as nitric oxide or carbon monoxide (7,8) or endogenous opioids.…”
