Role of the brain melanocortins in blood pressure regulation

Abstract: Melanocortins play an important role in regulating blood pressure (BP) and sympathetic nervous system (SNS) activity as well as energy balance, glucose and other metabolic functions in humans and experimental animals. In experimental models of hypertension with high SNS activity, blockade of the melanocortin-4 receptor (MC4R) reduces BP despite causing marked hyperphagia and obesity. Activation of the central nervous system (CNS) pro-opiomelanocortin (POMC)–MC4R pathway appears to be an important link between … Show more

“…Brain melanocortins.-A key CNS pathway that links adipokines and hormones with BP control is the brain POMC-melanocortin 4 receptor (MC4R) system 6,189,190 . In the ARC and brainstem, POMC-expressing neurons project to second-order neurons involved in BP regulation (for example, in the paraventricular nucleus (PVN)), where their axons release amelanocyte-stimulating hormone (aMSH), the endogenous ligand for MC4R 191 (FIG.…”

“…Increased sympathetic activity caused by hyperinsulinemia has been shown to decrease when melanocortin receptors are antagonized. 13,14 Adipose tissue produces pro-inflammatory cytokines such as tumor necrosis factor-α and interleukin-6. They can induced insülin resistance and promote endothelial dysfunction.…”

“…In addition to controlling feeding behavior, the ARC also regulates energy expenditure. For example, POMC/CART neurons decrease food intake and increase energy expenditure through the activation of MC3R or MC4R via the release of α-MSH [39, 40]. Conversely, AgRP/NPY neurons promote food intake and decrease energy expenditure [41].…”
