Role of the Interaction of Tumor Necrosis Factor-α and Tumor Necrosis Factor Receptors 1 and 2 in Bone-Related Cells

Abstract: Tumor necrosis factor-α (TNF-α) is a pleiotropic cytokine expressed by macrophages, monocytes, and T cells, and its expression is triggered by the immune system in response to pathogens and their products, such as endotoxins. TNF-α plays an important role in host defense by inducing inflammatory reactions such as phagocytes and cytocidal systems activation. TNF-α also plays an important role in bone metabolism and is associated with inflammatory bone diseases. TNF-α binds to two cell surface receptors, the 55k… Show more

“…The overall RSD value of 2.4% obtained for the current signals measured for the individual mixing of other cytokines with TNF-α and all together, with a maximum relative signal variation of 5%, confirmed that the selective detection of TNF-α cytokine could be achieved in the multicytokine sample matrix (Figure Biv). These findings were in agreement with early reports on cytokine–receptor interactions. , …”

“…These findings were in agreement with early reports on cytokine−receptor interactions. 33,34 The use of the specific detection antibody and standard HRP-TMB system enhanced the sensitivity of the biomembrane sensor. The analytical performance of the cell membrane-based bioelectronic chip for the sensitive detection of various TNF-α concentrations in the presence of IL-6 cytokine was also studied.…”

“…For example, a pro-inflammatory cytokine, TNF-alpha, induced the sclerostin expression in osteoblast and osteocyte cells ( 53 55 ). Moreover, TNF-alpha contributes to osteoporosis, promoting RANKL-induced osteoclast formation ( 56 , 57 ). In GD, the elevation of TNF-alpha is associated with macrophage and T-cell activations ( 58 60 ).…”

“…One of the ways to investigate the anti‐inflammatory effect of MSCs on HCC xenografts is by determining the expression of various inflammatory mediators such as IL‐1α, IL‐2, IL‐4, IL‐8, IL‐10, TNF‐α and transforming growth factor‐β (TGF‐β) . In this study, we chose to investigate the expression of IL‐1α, IL‐10 and TNF‐α, due to their key role in immune regulation, inflammatory responses, tissue remodelling, cell motility, cell cycle and apoptosis (Malik & Kanneganti, 2018 and Kitaura et al., 2022).…”
