Role of the small GTPase activating protein IQGAP1 in collagen phagocytosis

Abstract: Many adult connective tissues undergo continuous remodeling to maintain matrix homeostasis. Physiological remodeling involves the degradation of collagen fibers by the intracellular cathepsin-dependent phagocytic pathway. We considered that a multidomain, small GTPase activating protein, IQGAP1, which is involved in the generation of cell extensions, is required for collagen phagocytosis, possibly arising from its interactions with cdc42 and the actin-binding protein Flightless I (FliI). We examined the role o… Show more

“…MRC2-mediated collagen uptake occurs via receptor-mediated endocytosis. Collagen uptake can also occur via phagocytosis 24 or macropinocytosis 25 , and impairments in any of these processes due to gene silencing could potentially be detected in our screens. We therefore compared the enrichment of these collagen-related GO terms in our collagen uptake screens with data from previously published CRISPR screens of phagocytosis of other cargo 26 (Fig.…”

“…IQGAP1 is a ubiquitously expressed scaffold protein involved in various vital cellular functions ( 4 , 5 ). IQGAP1 has been postulated to maintain matrix homeostasis via collagen phagocytosis in physiological tissue remodeling ( 26 ). Furthermore, IQGAP1 appears to be an exceptionally attractive therapeutic target because it acts as a hub for signaling pathways involved in cancer progression ( 20 , 27 29 ).…”
