Roles and regulation of Thy‐1, a context‐dependent modulator of cell phenotype

Leukocyte recruitment in response to inflammatory signals is governed, in part, by binding to Thy-1 (CD90) on activated endothelial cells (EC). In this study, we characterized the adhesion G-protein coupled receptor CD97, present on peripheral myeloid cells, as a novel interacting partner for Thy-1. CD97 was upregulated on polymorphonuclear cells (PMNC) of patients with psoriasis. In psoriatic skin lesions, CD97+ myeloid cells colocalized with Thy-1+ EC of small vessels in microabscesses, suggesting an interaction between CD97 and Thy-1 that was further examined by adhesion and protein-binding assays. PMNC and cell lines stably overexpressing CD97 adhered specifically to Thy-1+–activated human dermal EC, Thy-1+ CHO cells, and immobilized Thy-1 protein. Binding of the CD97+ CHO clones correlated with their CD97 expression level. Soluble CD97 bound specifically to immobilized Thy-1 protein, as well as Thy-1+–activated EC and CHO cells. In all assays, cellular adhesion or protein binding was blocked partially by CD97 and Thy-1–blocking mAb. Our data suggested that CD97 interacts via its stalk with Thy-1 because mAb directed to the stalk of CD97 showed stronger blocking compared with mAb to its epidermal growth factor-like domains, and binding was calcium independent. Moreover, soluble CD97 without the stalk and soluble EMR2, containing highly homologous epidermal growth factor-like domains but a different stalk, failed to bind. In summary, binding of leukocytes to activated endothelium mediated by the interaction of CD97 with Thy-1 is involved in firm adhesion of PMNC during inflammation and may play a role in the regulation of leukocyte trafficking to inflammatory sites.

PURPOSE. Thy-1 is a marker of retinal ganglion cell (RGC) differentiation. Optic nerve injury triggers reduction of Thy-1 promoter activation followed by retinal ganglion cell (RGC) death. This study determined whether MS-275, an inhibitor of the histone deacetylases 1 and 3, can inhibit these changes.METHODS. Mice expressing cyan fluorescent protein (CFP) under control of the Thy-1 promoter received MS-275 (subcutaneous) or vehicle three times per week starting 1 week before optic nerve crush and continuing for 6 weeks. The same retinal area was imaged using the blue-light confocal scanning laser ophthalmoscope before and after optic nerve crush every week, and fluorescent spots were counted manually. The eyes were then processed for histopathologic analysis.RESULTS. The mean proportions of fluorescent retinal neurons remaining in the vehicle group following optic nerve crush were 36 6 8, 18 6 6, 13 6 10, 12 6 4, 13 6 5, and 13 6 5% at weeks 1 through 6, respectively (n ¼ 6). In contrast, the mean proportions of fluorescent retinal neurons remaining in the group treated with MS-275 were 59 6 19, 39 6 11, 34 6 12, 33 6 15, 32 6 13, and 27 6 15% at weeks 1 through 6, respectively (n ¼ 7, P < 0.05 at weeks 1 through 5). Rate analysis showed that MS-275 slowed the rate of loss during the first 2 weeks by 23% (P < 0.05) and subsequently was similar. Histopathologic analysis revealed 27 6 13% greater ganglion cell layer (GCL) neurons in the eyes from mice that received MS-275 treatment (P < 0.02).CONCLUSIONS. These results indicate that treatment with MS-275 protects against the loss of RGC differentiation and promotes RGC survival following optic nerve injury. (Invest Ophthalmol Vis Sci. 2013;54:96-102)

Na primeira, verificar se existem células-tronco mesenquimais derivadas da curetagem do alvéolo dental humano após extrações dentais. Na segunda, analisar o nicho de células-tronco nas polpas dentais de camundongos. Em comum, as duas análises se basearam no uso de marcadores previamente utilizados na literatura para o estudo de células-tronco. Como não existem marcadores únicos e específicos para a identificação dessas células, diferentes combinações desses marcadores entre si e de técnicas laboratoriais foram empregadas. No primeiro estudo, após o isolamento das células, deu-se especial atenção à sua caracterização, que foi realizada através de ensaios que avaliaram propriedades e comportamentos que sabidamente ocorrem em células-tronco. AGRADECIMENTOSJá na segunda parte desse estudo, utilizamos a polpa dental de camundongos para a análise in vivo de nichos. Em camundongos, a localização do nicho responsável pelo crescimento continuo dos incisivos é conhecida. De uma maneira geral, nossos resultados demonstram que: 1) É possível isolar células-tronco/progenitoras a partir de tecidos curetados de alvéolo dental após extrações dentárias. Esse fato é inédito e abre novas perspectivas em termos de oportunidade de coleta e futura aplicação clínica. 2) É possível observar a diversidade populacional nas células que formam o nicho de células-tronco em polpa dental de camundongos, e através de uma organização hierárquica, propusemos um modelo para este nicho. Esse modelo servirá de base para estudos subsequentes que visem avaliar o comportamento das células que formam o nicho, quando isoladas das demais, e abrirá possibilidade para análises em outros tecidos dentais e não dentais.Palavras-chave: Células-tronco dentais. Nicho de células-tronco There is great interest in the study of stem cells due to their ability to produce mature cells of different lineages that participate actively in the process of homeostasis, injury response, regeneration and tissue repair. The dental pulp is the most studied tissue in dentistry, but several studies have shown the presence of these cells in the periodontal tissue region. It is important to note that depending on their origin, these cells exhibit different behaviours, especially in regard to in vivo transplantation.
