Rspo1/Wnt signaling promotes angiogenesis via Vegfc/Vegfr3

Abstract: SUMMARYHere, we show that a novel Rspo1-Wnt-Vegfc-Vegfr3 signaling pathway plays an essential role in developmental angiogenesis. A mutation in R-spondin1 (rspo1), a Wnt signaling regulator, was uncovered during a forward-genetic screen for angiogenesisdeficient mutants in the zebrafish. Embryos lacking rspo1 or the proposed rspo1 receptor kremen form primary vessels by vasculogenesis, but are defective in subsequent angiogenesis. Endothelial cell-autonomous inhibition of canonical Wnt signaling also blocks an… Show more

“…Indeed, vegfc is expressed in ISVs as they sprout from the dorsal aorta and mosaic analysis demonstrates that vegfc-deficient endothelial cells are less likely to contribute to the tip cell position in ISVs. These results are consistent with the ISV growth defects observed in rspo1 mutant zebrafish, which lack endothelial vegfc expression, and can be rescued with an endothelial cell-specific vegfc transgene (Gore et al, 2011). Interestingly, ISV tip cells in rspo1 mutant embryos displayed significantly fewer filopodia, consistent with our observation that Vegfc contributes to filopodia stability.…”

“…Rspo1 has also been shown to be involved in mammary gland formation in mice and angiogenesis in zebrafish. In addition to its different roles in embryogenesis, this gene can also promote organ homeostasis, as evidenced by its role in b-cell neogenesis in the pancreas (Gore et al 2011, Yoon & Lee 2012.…”

“…In zebrafish, primary angiogenesis occurs when arterial sprouts emerge from the dorsal aorta (DA) from ∼22 hpf to give rise to the intersegmental arteries (aISV) in a process dependent on Kdr, Kdrl and Vegfa (Bahary et al, 2007;Covassin et al, 2006;Nasevicius et al, 2000;Olsson et al, 2006). During primary angiogenesis, blood vessel sprouting is led by tip cells, which express high levels of Flt4 (Vegfr3 in mammals) and display active Erk signaling (Gore et al, 2011;Shin et al, 2016a). In zebrafish, whole-genome duplication has resulted in the presence of two Vegfr2 paralogs, Kdr and Kdrl (Bussmann et al, 2008;Covassin et al, 2006).…”
