S-Adenosyl Methionine and Transmethylation Pathways in Neuropsychiatric Diseases Throughout Life

Abstract: S-Adenosyl methionine (SAMe), as a major methyl donor, exerts its influence on central nervous system function through cellular transmethylation pathways, including the methylation of DNA, histones, protein phosphatase 2A, and several catecholamine moieties. Based on available evidence, this review focuses on the lifelong range of severe neuropsychiatric and neurodegenerative diseases and their associated neuropathologies, which have been linked to the deficiency/load of SAMe production or/and the disturbance … Show more

“…Deficits in RELN are associated with illnesses like schizophrenia, autism, epilepsy, and others. While glutamic and aspartic acids are excitatory neurotransmitters that help long-term potentiation (LTP), GABA is inhibitory that in mature neurons curb impulsive and aggressive behaviors ( Box 1 ) [ 100 , 101 , 102 , 103 , 104 ].…”

“…SAMe is the most important methyl donor in mammalian cells, where it transfers a methyl group to acceptor molecules, such as DNA, proteins, and lipids, thus modifying their structure and function [ 5 ]. Many reports have shown that SAMe treatment causes the hypermethylation of DNA, which is typically associated with the silencing of gene expression [ 6 ]. For this reason, SAMe has been proposed for use in cancer therapy to reduce tumor development, growth, and metastasis.…”

“…Moreover, vitamin B12 injections represent the only proposed treatment in cases of specific diseases, such as rare genetic mutations in B12 metabolism [6][7][8]. Nevertheless, except for the efficient correction of the methyl donors (B-vitamins) deficiency during pregnancy, numerous cases of B9/B12 metabolism deregulation during the first years of life still lead to long lasting neurologic and behavioral dysfunction [9][10][11].…”
