Saccharopolyspora flava sp. nov. and Saccharopolyspora thermophila sp.nov., novel actinomycetes from soil.

Abstract: The generic position of two aerobic, Gram-positive, non-acid-alcohol-fast actinomycetes was established following the isolation of their PCR-amplified 16S rRNA genes and alignment of the resultant sequences with the corresponding sequences from representatives of the families Actinosynnemataceae and Pseudonocardiaceae. The assignment of the organisms to the genus Saccharopolyspora was strongly supported by chemotaxonomic and morphological data. The strains were distinguished both from one another and from repr… Show more

“…Some sequences were obtained from the public sequence databanks, but the vast majority were determined and deposited during the current study. DNA was isolated and approximately 1320 almost-complete 16S rRNA gene sequences were determined in the various collaborating laboratories using previously described methods (Chun and Goodfellow 1995;Goodfellow et al 2007;Kim et al 1996;Kumar and Goodfellow 2008;Labeda and Kroppenstedt 2000;Labeda et al 2009;Lu et al 2001;Park et al 2005).…”

“…PCR amplification of the 16S rDNA was carried out as described by Kim et al (1996) and the resultant PCR products were purified by using the Wizard PCR purification system (Promega), following the manufacturer's instructions. The purified products were sequenced directly using a Taq Dye Deoxy Terminator cycle sequencing kit (Applied Biosystems) and two universal primers described previously (Lu et al, 2001). Sequence gel electrophoresis was carried out and the nucleotide sequence was obtained automatically by using an Applied Biosystems DNA sequencer (model 373A) and software provided by the manufacturer.…”

“…The genus Saccharopolyspora was proposed by [7] originally isolated from spontaneously heated sugarcane bagasse. Representatives of these species form a distinct phyletic line within the evolutionary radiation encompassed by the family Pseudonocardiaceae can be distinguished by using phenotypic properties [8] . Members of the genus Saccharopolyspora are characterized by aerobic, extensively branched substrate hyphae that fragment into rod-shaped elements and aerial hyphae that segment into bead-like chains of spores.…”
