Safety and pharmacokinetic profile of fixed-dose ivermectin with an innovative 18mg tablet in healthy adult volunteers

Abstract: Ivermectin is a pivotal drug for the control of onchocerciasis and lymphatic filariasis, which is increasingly identified as a useful drug for the control of other Neglected Tropical Diseases. Its role in the treatment of soil transmitted helminthiasis through improved efficacy against Trichuris trichiura in combination with other anthelmintics might accelerate the progress towards breaking transmission. Ivermectin is a derivative of Avermectin B1, and consists of an 80:20 mixture of the equipotent homologous … Show more

“…This would be expected to give overall shorter elimination half-lives and lower peak concentrations as compared to our data. Mean elimination half-lives reported in literature following single oral doses of 200 μg kg À1 ivermectin range from 25 to 80.6 h, placing our estimate at the upper end [11,32,33]. The mean peak concentrations in these studies, whose weight-based dose is comparable to the mean dose of 181 μg kg À1 in our fixed-dose regimen, were consistently lower (43.2-50.1 ng ml À1 ).…”

“…In the last years, high doses of IVM have been evaluated for the treatment of soil-transmitted helminths [7][8][9][10] and as a new vector control tool to reduce malaria transmission in malaria endemic areas [11]. Recent studies have evaluated doses up to 800 μg/kg, given in single dose or three consecutive days [9,11,12], showing a good safety profile both in adult and paediatric populations.…”

“…Ivermectin (IVM) is the 22,23‐dihydro derivative of avermectin B1 and is an important member of the avermectin family of 16‐membered macrocyclic lactones evolve from the actinomycete Streptomyces avermectinius. IVM was originally used as a veterinary drug and was very effective for a variety of parasites, such as gastrointestinal roundworms, lungworms and mites …”
