Salibaculum halophilum gen. nov., sp. nov. and Salibaculum griseiflavum sp. nov., in the family Rhodobacteraceae

Abstract: Two Gram-stain-negative, moderately halophilic, non-motile, rod-shaped, pale yellow, and aerobic strains, designated WDS1C4T and WDS4C29T, were isolated from a marine solar saltern in Weihai, Shandong Province, PR China. Growth of strain WDS1C4T occurred at 10–45 °C (optimum, 37 °C), with 4–16 % (w/v) NaCl (optimum, 8 %) and at pH 6.5–9.0 (optimum, pH 7.5). Growth of strain WDS4C29T occurred at 10–45 °C (optimum, 40 °C), with 2–18 % (w/v) NaCl (optimum, 6 %) and at pH 6.5–9.0 (optimum, pH 7.5). Q-10 was the so… Show more

“…Expression of the sulphur-oxidizing (SOX) system was tested by using a medium (0.5 % peptone, 0.1 % yeast extract and 0.03 % sodium pyruvate), prepared with artificial seawater (MgCl 2 0.23 %, MgSO 4 0.32 %, CaCl 2 0.12 %, KCl 0.07 % and NaS 2 O 4 0.5 %, all w/v). S 2 O 3 2− and SO 4 2− were detected by FeCl 3 and BaCl 2 solutions at 0 and 7 days [ 47 ].…”

“…Genomic DNA of the two strains was extracted and purified using a Bacterial Genomic DNA Mini Kit (Takara Bio) according to the manufacturer's recommendations. The 16S rRNA gene of both strains was amplified by PCR using universal primers 27F and 1492R [7]. On the basis of pairwise comparisons of the 16S rRNA gene sequences using the EzBioCloud (www.ezbiocloud.net) [8] and NCBI [National Centre for Biotechnology Information (nih.gov)] data libraries, F47161 T and F64183 T had high similarity in the 16S rRNA gene sequences with A. sinensis JCM 19789 T (97.5 % and 98.4 %, respectively) and A. viscosa JCM 18497 T (97.5 % and 97.4 %, respectively).…”

“…The monomeric SoxB, which was defined as S-sulfosulfanyl-l-cysteine sulfohydrolas, interacted with the SoxYZ complex and was believed to function as a sulphate thiohydrolase [21]. Strain N1Y112 T and P. granulatum JCM 30316 T were incubated using marine broth (MB) prepared with each strains' optimum salinities and modified artificial seawater (0.67 % Na 2 S 2 O 3 , 0.48 % MgCl 2 , 0.12 % CaCl 2 , 0.07 % KCl and 0.02 % NaHCO 3 , all w/v [22]). The sulphate in the culture was detected by FeCl 3 and BaCl 2 solution at 0 and 7 days, and the results showed that sulphate produced from thiosulhate only in the culture medium of strain N1Y112 T (Fig.…”
