Salidroside attenuates neuroinflammation and improves functional recovery after spinal cord injury through microglia polarization regulation

Abstract: Spinal cord injury (SCI) is a severe neurological disease; however, few drugs have been proved to treat SCI effectively. Neuroinflammation is the major pathogenesis of SCI secondary injury and considered to be the therapeutic target of SCI. Salidroside (Sal) has been reported to exert anti‐inflammatory effects in airway, adipose and myocardial tissue; however, the role of Sal in SCI therapeutics has not been clarified. In this study, we showed that Sal could improve the functional recovery of spinal cord in ra… Show more

“…21,22 Based on the activation of BRD4 after the SCI described above and previous studies about the pro-inflammation property of BRD4, we decided to investigate the effects of BRD4 inhibition on inflammatory response after SCI. 17 Lipopolysaccharide (LPS) is a well-recognized inflammatory response inducer in microglia, thus we used it to trigger inflammatory response in HAPI microglia cells.…”

“…In contrast, M2 microglia/macrophages upregulate anti‐inflammatory or reparative factors, which protect the neural tissues (Alizadeh, Santhosh, Kataria, Gounni, & Karimi‐Abdolrezaee, ). Because M1 microglia/macrophages predominate in the spinal cord after acute SCI, regulating macrophage polarization may be a useful strategy for improving functional recovery (Wang et al, ). In our study, expression levels of iNOS and CD16 were increased in the spinal cord after SCI, as compared to levels in sham‐operated rats.…”

“…Like melatonin, salidroside can also promote functional recovery and ameliorate neuroinflammation after SCI by inducing microglia/macrophages to switch from the M1 to the M2 state. The salidroside‐induced phenotype switch is modulated by AMPK‐/mTOR‐mediated autophagic flux stimulation (Wang et al, ). Studies also have shown that hemopexin may attenuate SCI by regulating microglial/macrophages polarization via the LRP‐1 receptor (Han et al, ).…”
