Sample station modification providing on‐line reagent addition and reduced sample transit time for flow cytometers

Abstract: A flow cytometer was equipped with a modified sample station to facilitate online addition of mediators to the sample and reduce the time of delivery of the sample to the interrogation point. The ready availability of materials and straightforward nature of this design make modifications simple and facilitate measurements of cellular activation. Parameters such as pH, membrane fluidity, and calcium mobilization are easily measured in this system, because detection can be made less than 4 s after addition of me… Show more

“…To overcome the disadvantage typical for FCM as an off-line analyser system a number of flow injection flow cytometry (FI-FCM) systems for dyeing purposes have been designed (Omann et al, 1985;Pennings et al, 1987;Kelley, 1989). Ruzicka and Lindberg (1992;Lindberg et al, 1993Lindberg et al, , 1994 were the first using FI as an interface between a bioreactor and a FCM.…”

“…As mentioned FACS is one of the available methods for measurement of population distribution and has been exploited in biotechnology (Rieseberg et al, 2001). To overcome this disadvantage of FACS as an off-line analyser, a number of flow injection (FI) flow cytometer systems have been designed for the determination of proteins (Kelley, 1989). Ruzicka and Lindberg (1992) were the first to utilise FI as an interface between a bioreactor and FACS.…”

“…This problem was illustrated in a paper by Pennings et al (10) in which the authors, in a measurement of DNA and RNA using acridine orange, observed a continuous decrease in fluorescence intensity during the measurement in cases in which a 'This project is supported by the National Institutes of Health through grant no.
