SarS, a SarA Homolog Repressible by agr , Is an Activator of Protein A Synthesis in Staphylococcus aureus

Abstract: The expression of protein A (spa) is repressed by global regulatory loci sarA and agr. Although SarA may directly bind to the spa promoter to downregulate spa expression, the mechanism by which agr represses spa expression is not clearly understood. In searching for SarA homologs in the partially released genome, we found a SarA homolog, encoding a 250-amino-acid protein designated SarS, upstream of the spa gene. The expression of sarS was almost undetectable in parental strain RN6390 but was highly expressed … Show more

“…SarS has been reported to function as an activator of spa (surface protein A) expression (Cheung et al, 2001). This is consistent with the observation that expression of spa was also dramatically increased in UAMS-1 relative to RN6390 (Table 3).…”

“…Expression of both sarS and spa is repressed by agr (Cheung et al, 2001). The fact that agr is expressed at lower levels in UAMS-1 and other clinical isolates, relative to RN6390, may therefore explain why sarS and spa are produced at elevated levels in these strains.…”

“…2C). This feature may possibly explain how SarS, a 250-residue protein homologous to SarA (26), may function as a heterodimer-like monomer because it contains two sections, each highly similar to the SarR module and 125 residues long (26). The entire dimer can be described as three individual subdomain structures (Fig.…”

“…Perhaps the most consistent transcriptional change observed for VISA strains is the downregulation of the spa gene (74,122,169,199,201,251,303). A possibly incomplete list of genes where the gene product has been shown to affect the transcription of spa includes agr (76,343), sarA (51,76,343), sarS (50), arlSR (85,86), sarT (304), srrAB (262), tcaR (200), mgrA (138), rot (240), ccpA (307), and sarZ (342). Potentially, a mutation in any of these regulatory genes could simultaneously increase vancomycin resistance and decrease protein A production.…”
