SARS-CoV-2 detection with aptamer-functionalized gold nanoparticles

Abstract: A rapid detection test for SARS-CoV-2 is urgently required to monitor virus spread and containment. Here, we describe a test that uses nanoprobes, which are gold nanoparticles functionalized with an aptamer specific to the spike membrane protein of SARS-CoV-2. An enzyme-linked immunosorbent assay confirms aptamer binding with the spike protein on gold surfaces. Protein recognition occurs by adding a coagulant, where nanoprobes with no bound protein agglomerate while those with sufficient bound protein do not. … Show more

“… 51 In another work, the use of aptamer-functionalized gold nanoparticles was reported, achieving a detection limit of 10 nM, which is equivalent to 3540 genome copies/μL of inactivated SARS-CoV-2. 67 Using a 44-mer G-quadruplex-forming DNA aptamer they selected against the spike trimer antigen of SARS-CoV-2, Gupta et al achieved a detection limit of 2 nM. 52 They used this aptamer for the analysis of 232 NP samples, achieving a highly discriminatory response between SARS-CoV-2-infected individuals from the noninfected ones with a sensitivity and specificity of 91 and 98%, respectively.…”

“…Another study used antigen-coated Au NPs through the principle of plasmon resonance to assist in the detection of the N protein of SARS-CoV-2 [ 213 ]. Au NPs could be also used as nanoprobes to detect the S protein of SARS-CoV-2 [ 214 ].…”

“…According to statistics, compared with a single PCR test, the positive detection rate is significantly increased from 51.9 % to 98.6 % by combining the PCR with the ELISA assay for each patient [ 116 ]. Srivatsa et al used aptamer-functionalized gold nanoparticles to identify SARS-CoV-2 spike proteins, limit of detection can up to 3540 genome copies/μl [ 117 ]. However, the cross-reactivity and low antibodies titers are among common factors that limited the detection efficacy of ELISA.…”
