Scheduled simple production method of pseudopregnant female mice for embryo transfer using the luteinizing hormone-releasing hormone agonist

Abstract: The use of mice as experimental animal models has been a practice since the development of genetically engineered mouse models (GEMMs) in the early 1980s. New technologies, including genome editing, have helped in the time- and cost-efficient generation of GEMMs. However, methods for preparing pseudopregnant females, essential for the generation of GEMMs, remain less advanced. This study proposes a new method to achieve simple production of pseudopregnant female mice using a luteinizing hormone-releasing hormo… Show more

“…One possible solution to this issue is using a deep learning-based method (Sano et al, 2020). In addition, combining our method and some studies on synchronizing the estrus cycle by hormone administration may be effective in producing pseudopregnancy (Hasegawa et al, 2017;Sari et al, 2022). Another approach for enhancing pregnancy rates involves monitoring ovulation and the thickness of the fallopian tubes or uterus.…”

“…Fertile and sexually active male rats were introduced on day three after administration of RICOM 1013-J into each group (ratio of 3 females to 1 male per cage) till the end of experiments for mating according to the methods of [10]. Mating was confirmed by the presence of a whitish vaginal plug with the presence of spermatozoa and is considered to be day '1' of conception [27,28]. The male rats were in contact with the female rats for the seven gestation periods of the study.…”

“…Then, 10 to 16-week-old C57BL/6 female mice purchased from BioLASCO Taiwan were used to induce pseudopregnancy and estrus synchronization was induced by administration of the luteinizing hormone releasing hormone agonist (LHRHa), as described in the previous study. 103 To efficiently synchronize the mating time for both pseudopregnant and superovulated female mice, the estrus synchronization was induced by the first i.p. injection of 20 μg of the LHRHa, namely [des-Gly 10 , D-Ala 6 ]-LH-RH ethylamide acetate salt hydrate (Sigma-Aldrich Product No.…”
