Sclerosing Polycystic Adenoma of Salivary Glands

Abstract: Sclerosing polycystic adenoma (SPA) is a rare salivary gland neoplasm originally thought to represent a non-neoplastic lesion. Recently we have encountered an index case of apocrine intraductal carcinoma of parotid gland of 62-year-old man with invasive salivary duct carcinoma component arising from SPA, a combination of tumor entities that has never been published so far. Here, we further explore the nature of SPA by evaluating 36 cases that were identified from the authors’ consultation files. The patients w… Show more

“…Apocrine IDC and high‐grade SDC were both components of a SPA case analyzed recently by Skálová and colleagues, 10 and all three of these components carried the same mutation in the PI3K/Akt pathway. The authors suggest that a close relationship between these three entities must exist, and SPA may represent a precursor lesion for the development of apocrine IDC and invasive SDC 6,10 …”

“…Fluorescence in situ hybridization (FISH) and next‐generation sequencing (NGS) were used to describe the SPA molecular profile. Mutations in PIK3CA gene were found in 16 cases, 9–11 PTEN in 10 cases, 9–11 HRAS in 4 cases, 10 and AKT1 in 2 cases 10 . Monoclonality of the cells was revealed in all female patients investigated using a PCR‐based analysis of patterns of X chromosome inactivation using human androgen receptor (HUMARA) locus 3 .…”
