DNA methylation of Th2 lineage determination genes at birth is associated with allergic outcomes in childhood

Abstract: Our findings provide further evidence of a developmental contribution to the risk of later allergic disorders and suggest that involvement of epigenetic mechanisms in childhood asthma is already demonstrable at birth.

“…Also, a relative hypermethylation of cg04256470 (688 bases upstream of CORT) was associated not needing oral corticosteroid in all cohorts. These are interesting, robust associations which may help to elucidate why some children with asthma do better than others and echo other studies …”

“…These are interesting, robust associations which may help to elucidate why some children with asthma do better than others and echo other studies. 3,4 Ballardini et al have assessed the size of the association between childhood atopic dermatitis and asthma with childhood behaviour. 5 They analysed data from the PROmotion of Breastfeeding Intervention Trial (PROBIT) cohort where participants were followed up to 6.5 years of age.…”

“…Finally, Clinical and Experimental Allergy has also included reports on sensitization patterns for rhinitis and asthma multimorbidity from the EGEA study, 67 an association between DNA methylation of Th2 lineage determinants of genes at birth and later allergic outcomes in the EpiGene consortium 68 The antigen-specific CD4+ Th2 T cell response is thought to be mediated by DC presentation of pollen allergens to naïve CD4+ T cells in the draining lymph nodes. 70,73 Critical steps for the DC-CD4+ type 2 cell polarization is the interaction of co-stimulatory molecules (CD28 and CD80/CD86; OX40L and OX40) between antigen-presenting cells (APCs) and T cells.…”

“…Barton et al have looked for possible associations between epigenetic changes around key genes at birth and the development of allergic disease. In the Southampton Women's Survey, they found that higher methylation of GATA3 CpGs ‐2211/‐2209 at birth was associated with less likelihood of asthma at age 3 and 6 (Figure ).…”
