PP2A‐B55 and its adapter proteins IER2 and IER5 regulate the activity of RB family proteins and the expression of cell cycle‐related genes

Abstract: The retinoblastoma (RB) tumour suppressor protein regulates cell proliferation, motility, differentiation and apoptosis. The phosphorylation state of RB is modulated by kinases and phosphatases, and RB exhibits phosphorylation‐sensitive interactions with E2F family transcription factors. Here, we characterize RB dephosphorylation by protein phosphatase 2A (PP2A). The growth factor‐inducible immediate early response (IER) proteins IER2 and IER5 possess an adapter‐like function in which IER proteins bind to both… Show more

“…New treatment options and combinations are therefore needed. Single-dose radiosurgery of melanoma brain metastases achieves 1-year local control rates of 86% [ 86 , 87 ] and comparable outcomes are achieved in larger metastases with hypofractionated radiosurgery delivered in three to five fractions [ 88 ]. Retrospective analyses suggest that the combination of stereotactic radiotherapy and immune checkpoint inhibitors achieves a survival benefit without increased toxicity compared to stereotactic radiotherapy or immune checkpoint inhibitors alone [ 89 , 90 , 91 , 92 , 93 , 94 , 95 , 96 , 97 ].…”

“…PARP1 stabilises IER5 protein and along with Ku70-IER5 promotes NHEJ [ 83 ]. IER5 also induces G2/M cell cycle arrest by inhibiting CDC25B transcription vis dephosphorylation by PP2A-B55 phosphatase [ 84 , 85 , 86 ]. Thus, IER5 appears to exert its function in a context-dependent manner involving its binding to various partners, but these diverse functions of IER5 may all contribute to stress resistance in both normal and cancer cells.…”
