RASSF 1A controls tissue stiffness and cancer stem‐like cells in lung adenocarcinoma

Abstract: Lung cancer remains the leading cause of cancer‐related death due to poor treatment responses and resistance arising from tumour heterogeneity. Here, we show that adverse prognosis associated with epigenetic silencing of the tumour suppressor RASSF1A is due to increased deposition of extracellular matrix (ECM), tumour stiffness and metastatic dissemination in vitro and in vivo. We find that lung cancer cells with RASSF1A promoter methylation display constitutive nuclear YAP1 accumulation and expression of prol… Show more

“…Further studies comparing CSC-related genes of cells cultured in 2D and Col-I-coated 2D are then needed. High Col-I density modestly impacts Nanog nuclear levels in colorectal carcinoma cells in comparison to low Col-I density (Pankova et al, 2019). As for MCF-7, our results are in line with Sox2 and Bmi1 transcripts, but not nuclear ERa, being increased in Col-I-enriched ER + murine breast tumors (Shea et al, 2018).…”

“…H&E staining is the gold standard for tumor diagnosis. With the development of biomechanics in the past two decades [46,47], the mechanical properties of tissues need to be investigated based on biomedical and physical parameters. In this study, we have assayed the changes in mechanical properties at the micro-nanometer level using AFM and determined the association between the TNM grade, metastasis, and stiffness of tumor samples.…”
