TDP‐43 and tau concurrence in the entorhinal subfields in primary age‐related tauopathy and preclinical Alzheimer's disease

Abstract: Phosphorylated tau (p‐tau) pathology correlates strongly with cognitive decline and is a pathological hallmark of Alzheimer's Disease (AD). In recent years, phosphorylated transactive response DNA‐binding protein (pTDP‐43) has emerged as a common comorbidity, found in up to 70% of all AD cases (Josephs et al., Acta Neuropathol, 131(4), 571–585; Josephs, Whitwell, et al., Acta Neuropathol, 127(6), 811–824). Current staging schemes for pTDP‐43 in AD and primary age‐related tauopathy (PART) track its progression … Show more

“…As shown in Figure 3e, we discovered that the volume and thickness of entorhinal cortex rank high position. It is to say, the entorhinal cortex plays a crucial role in early AD prediction, which supports previous findings in pathology [33].…”

“…6A). Cortically, we found significant negative correlations with age in the prefrontal cortex and insula, whilst the temporal and parahippocampal cortices did not yield significant correlation – which is consistent with findings from studies of cortical thickness [27,66]. The white matter is known to decline steadily after ∼35 years [64,65], and such negative correlation is also detected by NextBrain .…”

“…Given that the neuropathological changes in AD tend to originate in the EC 7 , 35 , this finding might imply that differences in EC rates of volume loss are more evident at younger ages preceding MCI onset. Recent studies have identified specific subfields in the EC affected by tau pathology in the preclinical stage—a stage that precedes MCI during which disease-related neuropathology has begun but clinical symptoms are yet to manifest 36 . There is also evidence that entorhinal dysfunction, that precedes neurodegeneration, begins in adults as young as 18–30 years 37 .…”
