WDFY 1 mediates TLR 3/4 signaling by recruiting  TRIF

Abstract: Toll-like receptors (TLRs) are pattern recognition receptors that sense a variety of pathogens, initiate innate immune responses, and direct adaptive immunity. All TLRs except TLR3 recruit the adaptor MyD88 to ultimately elicit inflammatory gene expression, whereas TLR3 and internalized TLR4 use TIR-domain-containing adaptor TRIF for the induction of type I interferon and inflammatory cytokines. Here, we identify the WD repeat and FYVE-domaincontaining protein WDFY1 as a crucial adaptor protein in the TLR3/4 s… Show more

“…All of WDFY family proteins contain WD40 repeats and WDFY1, 2 and 3 contain a FYVE domain, which is associated with vacuolar/lysosomal trafficking. WDFY1 and WDFY2 are medium size proteins (around 400 amino acids) and have been associated with endosomal pathway [11] and [12]. WDFY3 and WDFY4 are large proteins (>3000 amino acids) and contain PH and BEACH domains.…”

“…The most notably upregulated gene in the PBS-treated PAI-1 −/− myocardium is Wdfy1 , which encodes a critical mediator of the Toll-like receptor signaling pathway. 32 Also of interest, 2 genes encoding proteins that have been implicated in the negative regulation of the TGF-β signaling pathway, nuclear receptor 4a1 33 and Smad6, 34 are downregulated in the unstimulated PAI-1 −/− ventricle. Subacute treatment with AngII affected significantly more cardiac transcripts in the PAI-1 −/− versus WT (6005 versus 551).…”

“…For instance, Calm3 is the linear transcript of circRNA_41204 (one of the top 5 decreased circRNAs in intestinal I/R) and it encodes calmodulin (CaM), a ubiquitous Ca 2+ -binding protein that regulates a vast array of Ca 2+ -sensitive biological events, which play important roles in the pathogenesis of I/R injury [28,29]. Wdfy1 is the linear transcript of circRNA_18944 (one of the top 5 decreased circRNAs in iPoC), and its protein product was found to recruit the signaling adaptor protein TRIF to Toll-like receptors (TLRs) 3 and 4, thereby mediating the activation of NF-κB and induction of inflammatory cytokines [30]. Interestingly, the TLR signaling pathway was reported to be involved in the ischemic tolerance induced by preconditioning or postconditioning [31].…”
