Screening of Antibiotic Producing Actinomycetes for Antibiosis from Soil of Siraha, Nepal

Abstract: The increasing need of novel antibiotics has provided a pace for the search of antibiotics from actinomycetes. Primary and secondary screenings of antibiotic producing actinomycetes from the soil of Siraha (75-600 m) were performed. The minimum inhibitory concentration of the metabolites was determined against E. coli. Macroscopic, microscopic and biochemical characterization were performed for the identification of presumptive genera. Characterization of the antibacterial substances was done by TLC. Among 92 … Show more

“…Since the evaporation of crude filtrate was used to produce these metabolites, it was likely to have a high MIC value. Similar findings were also reported by Gurung, Sherpa, Agrawal, Lekhak, et al (2009) and Sah et al (2017). TLC showed only one antibiotic solution created the spot, indicating the existence of a single compound.…”

“…Sah et al (2021) extracted antibacterial metabolites from the fermentation liquid using various organic solvents (n-butanol, chloroform, dichloromethane, ethyl acetate, and methanol), but only the ethyl acetate solvent was able to extract potent metabolites at a detectable level from the fermented broth. This could be attributed to the superior solubility of the metabolites in the ethyl acetate solvent (Sah and Lekhak 2017). The extracted ethyl acetate from Actinomycetes B.11 against A. niger and C. albicans, while the ethyl acetate extract from Actinomycetes B.17 only inhibits C. albicans.…”

“…Actinomycetes are Gram-positive bacteria that can live on a wide range of substrates; both the simple and complex ones. Several general bacterial isolation procedures can be used to isolate the actinomycetes, including serial dilution; pouring plates, streaking, and centrifugation (Majhi, 2018). In the soil samples, centrifugation followed by repeated dilutions can enhance the growth of actinomycetes.…”
