Screening ofChinese cabbage mutants produced by60Co γ‐ray mutagenesis of isolated microspore cultures

Abstract: Since the release of the Chinese cabbage genome sequence, increasing interest has focused on the functional analysis of unidentified genes in Chinese cabbage. Mutant analysis forms the basis of functional genomics research. To produce a variety of Chinese cabbage mutants in the same genetic background, buds containing late uninucleate spores from a doubled haploid line of the Chinese cabbage variety 'Fukuda 50' were irradiated with 60 Co c-rays at doses of 20, 40 and 60 Gy. Then, the treated microspores wer… Show more

“…In this study, we characterized the drm exhibiting a slow-growing, chlorophyll-deficient phenotype, which was screened by Huang et al ( 2014 ) and derived from the Chinese cabbage DH line “FT” by a combination of isolated microspore culture and radiation treatment ( 60 Co γ-rays). Compared with the wild-type “FT” line, the chlorophyll contents were significantly reduced in drm , and the chloroplast structure was abnormal.…”

“…The drm was screened by Huang et al ( 2014 ) and derived from Chinese cabbage DH line “FT” by a combination of isolated microspore culture and radiation treatment ( 60 Co γ-rays). This mutant was diploid as determined by flow cytometry and exhibited stable inheritance after multiple generations.…”

“…An etiolated mutant ( pem ) was obtained using the combination of 60 Co‐γ radiation and isolated microspore culture in Chinese cabbage (Huang et al ). The phenotype of the pem mutant was etiolated throughout the entire growth cycle of the plant.…”

“…This strain was characterized by yellow leaves and elongated hypocotyls [44]. The parent used for the segregating population development with pylm in this study was 'FT', a DH line derived from Chinese cabbage variety 'Fukuda 50' screened by Shenyang greenstar Chinese cabbage research institute (Shenyang, China), which exhibits folded green leaves [68]. The pylm was reciprocally crossed with 'FT' to produce the F 1 , F 2 , and BC 1 generations.…”
