Backgrounds: Pinostrobin has the potential activity as an anti-cancer. However, its activity is still lower than the anticancer drugs on the market. To increase its activity, pinostrobin derivatives have been synthesized, namely pinostrobin propionate and pinostrobin butyrate, which are predicted to have better activity and lower toxicity than pinostrobin after being tested by in silico approach. So the compound deserves to be tested for its anticancer activity and selectivity on normal cells. Objective: This study aims to determine the anticancer activity of pinostrobin propionate and pinostrobin butyrate against the T47D breast cancer cell line and its selectivity against the Vero cell line. Methods: The cytotoxicity test which is anticancer activity test and its selectivity on normal cell were carried out using the MTT(3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay. The cells used were breast cancer cell line T47D and normal Vero cells. The test results were analyzed using a microplate reader with a wavelength of 570 nm. Results: From the analysis of anticancer activity on T47D cells, the IC50 values of pinostrobin, pinostrobin propionate, and pinostrobin butyrate were 2.93, 0.57, and 0.40 mM, respectively. While the results of the cytotoxicity test on Vero cells obtained the CC50 value of pinostrobin, pinostrobin propionate, pinostrobin butyrate was 1.27, 0.94, and 0.89 mM, respectively. So the SI value of pinostrobin (SI=0.4) is smaller than its derivatives (SI=1.7 and 2.2). Meanwhile, pinostrobin butyrate is more selective than pinostrobin propionate. Conclusions: It can be concluded that pinostrobin propionate and pinostrobin butyrate compounds have greater activity and selectivity than pinostrobin so these compounds are promising to be further developed as anticancer candidates.

Burns refer to damage to the skin's surface caused by exposure to high temperatures, which can be due to factors such as oil, water, electricity, fire, sun exposure, and chemicals. Prompt and appropriate treatment is essential to prevent undesirable consequences. Thus, this study aimed to quantify mangiferin, a potential treatment for burns, in the bioactive fraction of mango leaves (Mangifera indica L.) and evaluate its effectiveness in healing burns.The methods employed included thin-layer chromatography (TLC)-densitometry with validation measures, including linearity, detection and quantification limits (LoD and LoQ), precision, accuracy, and quantification. The bioactive fraction was formulated in membranes at concentrations of 5%, 10%, and 15%. These membranes were applied to rabbits previously subjected to six wound burns, and the healing progress was monitored by measuring burn diameter using a vernier caliper every 3 days for a total of 21 days. Mangiferin, the active compound, was detected at a wavelength of 257 nm. Test results yielded a linearity equation, y = 76496x + 2935.7, with a correlation coefficient value of 0.9957, a detection limit of 2.01 µg/mL, a quantification limit of 6.07 µg/mL, a coefficient of variation ranging from 0.59% to 3.33%, and an accuracy range of 99.18% to 100.9%, with mangiferin levels at 208.31 µg/mL. The membrane preparations of the bioactive mangiferin fraction were evaluated on second-degree burns in rabbits, with concentrations of 10% and 15% showing the most effectiveness.

Objective: Maize (Zea mays L.) is a crop that has been widely cultivated in Indonesia. Using corn kernels on a large scale will produce much corn cob waste, usually unused. According to the literature search, corn cobs’ phytochemical studies and pharmacological activities still need to be improved. This study aims to determine the content of secondary metabolites (metabolite profiling) and their antityrosinase and anti-aging potential. Methods: Corn cobs were macerated with methanol and fractionated with n-hexane, ethyl acetate, and butanol. The phytochemical profiling approach of the methanol extract was performed by liquid chromatography-mass spectra (LC-MS/MS). Anti-tyrosinase and anti-aging bioactivity were evaluated by thin layer chromatography (TLC)-bioautography and IC50 spectrophotometrically. Results: The evaluation results show that the butanol fraction leads to a potential value (IC50 99.92 µg/ml). Several compounds, especially flavonoid compounds (including catechin; kaempferol 3-arabinofuranoside 7-rhamnoside; 6,8-Di-C-beta-D-arabino pyranosyl apigenin; 5,7-Dihydroxy-8,4’-dimethoxyisoflavone) were identified by LC-MS/MS by comparing the molecular mass of MS/MS data with literature data. Conclusion: Based on this study, it can be concluded that butanol is the fraction that most actively inhibits tyrosinase, elastase, and collagenase enzymes, which means it potentially becomes a new anti-aging candidate.
