Biofungicides based on plant oils have some advantages compared to chemical fungicides, especially considering their harmful effect on the environment. Twenty-two essential oils from Germany and Albania were assayed for inhibitory and fungicidal activity against Verticillium dahliae Klebahn, the causal agent of Verticillium wilt of pepper, using the macrodilution fumigant method. The concentrations of oils obtained in the vapour phase were: 0.02, 0.04, 0.08, 0.16 and 0.32 μl ml-1 with determined minimum inhibitory and fungicidal concentrations. The strongest activity was shown by two samples of mint oil (Mentha piperita L.) at 0.02 μl ml-1 of air, both from Germany and Albania, followed by plant essential oils of eucalyptus (Eucalyptus globulus Labilardie), black pine (Pinus nigra L.) and cade (Juniperus oxycedrus L.), and all of them were lethal to the pathogen. Nine oils: two samples of mint, cade, eucalyptus, black pine, lavender (Lavandula angustifolia Mill.), sage (Salvia officinalis L.) and rosemary (Rosmarinus officinalis L.) inhibited the growth of Verticillium dahliae, while five oils (two samples of mint, eucalyptus, black pine and cade) showed fungicidal effects on the pathogen. These results showed that mint, eucalyptus, black pine and cade essential oils have a potential for further in vivo experiments against Verticillium dahliae.

RESUMO: Este estudo objetivou avaliar a ação antimicrobiana e antisséptica do extrato etanólico bruto da folha da Hymenaea martiana (Jatobá). O estudo foi realizado no Laboratório de Microbiologia e Imunologia da UNIVASF, na cidade de Petrolina-PE. Os extratos foram preparados utilizando diferentes diluentes, sendo estes: álcool etílico absoluto e a água destilada. Em seguida, foi empregada a técnica da Concentração Inibitória Mínima (MIC) e da Concentração Bactericida Mínima (CBM). Todos os ensaios foram realizados em triplicata. A CBM média do extrato diluído em etanol foi de 358μg/μL e do extrato diluído em água destilada foi igual a 520,82μg/mL. Não houve diferença (P<0,05) quanto à inibição bacteriana para o extrato diluído em álcool etílico absoluto ou água destilada autoclavada. Ao comparar a atividade do extrato diluído em álcool etílico absoluto e a relação com a presença do gene blaZ, observou-se que os isolados negativos para o gene pesquisado apresentaram uma CBM igual a 412,3μg/mL, e, quando comparadas aos que foram positivos para o gene blaZ, que foi de 308,80μg/mL, contudo sem diferença estatística. Quanto à inibição das bactérias utilizando extrato aquoso, a atividade foi igual para as bactérias com ou sem o gene (520,82μg/mL). Desse modo, tanto o extrato diluído em álcool etílico absoluto quanto em agua destilada autoclavada demonstrou atividade antimicrobiana, sugerindo que ocorreu extração de substâncias bioativas. Em relação ao potencial antisséptico, H. martiana teve ação pareada com o cloro, contudo aquele agiu mais rapidamente, enquanto o cloro agiu de modo ideal uma hora após a aplicação; ambos os resultados destacam que o extrato etanólico bruto das folhas de H. martiana possui potencial de combate à proliferação de bactérias ambientais e infecciosas, surgindo como uma forma de prevenir a mastite.

Background and aims The olive root endophyte Pseudomonas fluorescens PICF7 is an effective biocontrol agent of Verticillium wilt of olive (VWO). Colonization of olive roots either by strain PICF7 or by Verticillium dahliae triggers differential systemic transcriptomic responses, many of them related with defense-related genes. The aims were to develop an olive split-root system for assessing VWO development and biocontrol effectiveness of strain PICF7 in plants with a divided root architecture, and Revised version without track changes Click here to download Revised version without track changes renamed_090a5.docx for evaluating systemic defense responses during this tripartite interaction when strain PICF7 and V. dahliae are spatially separated. Methods An olive split-root system was generated and disease development, biocontrol effectiveness and systemic genetic responses in these plants upon strain PICF7 and V. dahliae colonization were compared to those reported and observed in olive plants grown under standard conditions (single pots). Specific defense-related genes, previously identified during PICF7-and/or V. dahliae-olive root interactions were selected and their expression patterns assessed in above-ground tissues by real-time qPCR analyses. Results Symptoms of VWO developed similarly both in split-root and single-root plants. However, even though PICF7 triggered systemic defense responses in aerial tissues prior to the infection by V. dahliae, effective biocontrol was not observed under these experimental conditions. While most of studied genes showed similar expression patterns along time in both systems (i.e. split root and single pot), some of them (e.g. the caffeoyl-O-methyltransferase coding gene) varied depending on whether strain PICF7 and V. dahliae were spatially separated or shared the same compartment. Conclusions A successful split-root system was generated to investigate genetic events taking place during the tripartite interaction olive-V. dahliae-P. fluorescens PICF7. VWO biocontrol by strain PICF7 must rely on mechanisms other than induction of systemic resistance responses. The expression pattern of specific defense-related olive genes depended on whether or not the biocontrol agent and the pathogen share the same root/soil region.
