Seasonal genetic variation associated with population dynamics of a poecilogonous polychaete worm

Abstract: Poecilogonous species show variation in developmental mode, with larvae that differ both morphologically and ecologically. The spionid polychaete Pygospio elegans shows variation in developmental mode not only between populations, but also seasonally within populations. We investigated the consequences of this developmental polymorphism on the spatial and seasonal genetic structure of P. elegans at four sites in the Danish Isefjord‐Roskilde‐Fjord estuary at six time points, from March 2014 until February 2015.… Show more

“…DNA was extracted from whole host individuals using DNeasy Blood and Tissue Kit (Qiagen) following the manufacturer's protocol for animal tissue and stored in −20°C (Thonig et al, 2017), and DNA concentration was measured with Qubit 4.0 Fluorometer with 1X dsDNA HS Assay (Thermo Fisher Scientific). In the current study, a subset of those samples was analyzed with ddPCR (Table 1).…”

“…In this study, we examined marine apicomplexan infections, coinfections, and their associations with host genetic diversity in three natural populations of the polychaete worm, Pygospio elegans . We determined genetic diversity by genotyping individual hosts at seven neutrally evolving microsatellite loci (Thonig et al, 2017 ) and inspected infection patterns of an undescribed marosporidian, Rhytidocystis sp., and the gregarine species, Selenidium pygospionis , that live in symbiosis with the host. Our aim was to determine whether infection dynamics are associated with differences in population genetic diversity and heterozygosity among individual hosts.…”

“…It is also important to consider the context, and the life history of the focal species when assessing SGDCs. Since genetic diversity in species with short life spans can have seasonal fluctuations [ 68 ], spatial SGDCs based on such species could lead to non-significant results, no SGDC or even skewed conclusions, raising the question if the theory is appropriate for these species. SGDCs assessed using species with annual lifecycles on a temporal scale will, however, provide information about responses to seasonal environmental changes and population dynamics.…”
