Secretion of functional antibody and Fab fragment from yeast cells.

Abstract: We have constructed yeast strains that secrete functional mouse-human chimeric antibody and its Fab fragment into the culture medium. For chimeric whole antibody, cDNA copies of the chimeric light-chain and heavy-chain genes of an anti-tumor antibody were inserted into vectors containing the yeast phosphoglycerate kinase promoter, invertase signal sequence, and phosphoglycerate kinase polyadenylylation signal. Simultaneous expression ofthese genes in yeast resulted in secretion of properly folded and assembled… Show more

“…Although there were previous reports of secretion of fulllength antibody by yeast cells (11,32), the titers attained were much lower than those that we report here. We observed higher titers of secreted trastuzumab (up to 0.9 g/liter) than of Hu1D10 (0.2 g/liter).…”

“…Wood et al (32) showed that glycosylated and nonglycosylated forms of heavy chain were produced in yeast cells. Similarly, Horwitz et al (11) showed that heavy chain ␥ produced in yeast cells appeared as a doublet on reducing SDS-PAGE, although the glycosylation states of the two heavy-chain forms were not determined. We were able to produce an aglycosylated mutant form of trastuzumab by changing the Asn residue at position 297 to Gln, but the titers of this form of the secreted antibody were reduced.…”

“…Some reports have addressed the production of antibody by yeast, although in those studies, productivity was either low or not mentioned (15,20,25,30,44). For the production of antibodies in yeast, it will be necessary to increase productivity.…”

“…While an efficient ADCC activity was observed, recombinant IgG1 failed to induce CDC. 113 A 180-fold improvement in secretion was obtained by using a mutant of α mating factor 1 leader peptide and by co-expressing protein disulfide isomerase (PDI). 114 Evans et al recently reported the production at high yield (40-90 mg/l) of scFv fused to human serum albumin.…”
