Sedum takesimense Protects PC12 Cells against Corticosterone-Induced Neurotoxicity by Inhibiting Neural Apoptosis

Abstract: Neuronal cell death induced by chronic stress in the central nervous system is a cause of neurological dysfunction. We investigated the neuroprotective potential of a water extract of S. takesimense (WEST) against corticosterone-induced apoptosis in PC12 cells and the possible underlying mechanisms. Cells were pretreated with 50 µg/mL of WEST to evaluate its neuroprotective effect based on endoplasmic reticulum (ER) stress inhibition and mitochondrial function improvement. Pretreatment with WEST prevented cort… Show more

“…PC12 cells were derived from a pheochromocytoma cell line of rat adrenal medulla, and it is often used for in vitro studies of neurological diseases. CORT-treated PC12 cells are also often used to evaluate the effectiveness of antidepressant drugs [21][22][23][24]. We investigated the effects of crocin on the survival rate and neurons of CORT-induced PC12 cells.…”

“…This has followed with reductive nerve cell apoptosis and has shown treatment effects on anxiety and other mental disorders ( 101 ). The water extract of Sedum Takesimense (WEST) also has a similar effect ( 102 ) ( Table 1 and Figure 2 ).…”

“…When the body is stimulated by stressors, it sends signals to the hypothalamus to activate the HPA axis, leading to the synthesis and secretion of GC (Cort) in the adrenal cortex, which plays a crucial role in the regulation of various processes including metabolic and immune functions. 27,28 Serum CK-MB and Tn-Ⅰ are markers commonly used to diagnose myocardial injury, with CK-MB mainly found in the myocardium and detected as a result of myocardial necrosis, and Tn-Ⅰ, a protein unique to cardiac cells, whose increased levels in the blood also indicate myocardial damage. 29 It is often used as a diagnostic indicator of acute myocardial injury, and increased LDH activity is also indicative of myocardial cell damage.…”
