Selection of Media for Tissue and Cell Culture

Abstract: This chapter deals with the selection of growth media for the induction of callus and initiation of liquid suspension cultures from plants. Although the first attempts at initiating cultures of plant cells were made by the German botanist G. Haberlandt at the turn of this century, it has only been during the last three decades that rapid developments in plant cell, tissue, and organ culture have occurred. Since extensive work with microbial cultures was by then already underway, the first medium formulations u… Show more

“…NMR Measurements. 1 H and 19 F NMR spectra were recorded on a Varian Unity-500 spectrometer at 499.8 MHz for 1 H and 470.3 19 F NMR in deuteriochloroform, using tetramethylsilane ( 1 H) δ ) 0.0 and hexafluorobenzene ( 19 F) δ ) -162.9 as internal standards.…”

“…Theoretical background for an explanation of changes in 19 F NMR spectra is based on differences in shielding of the CF 3 moiety on the same side of the CF 3 -C-CO-CH plane, producing the deviation of the CF 3 group from this plane Final assignments of absolute configuration on chiral centers C-3 and C-4 were based on the following assumptions: (i) the cyclohexane ring is in the chair form; (ii) the bulky 2-hydroxy-2-propyl moiety is in the thermodynamically more stable equatorial position. 1 H and 19 F NMR data of measured MTPA esters are shown in Table 1 (Supporting Information). Comparing the 1b and 1c chemical shifts of protons H-2 and H-7 demonstrated an upfield shift (∆δ ) δ (R)-MTPAδ (S)-MTPA ), in the case of protons H-4, H-9, and H-10 and then a downfield shift.…”

“…1E, medium sugar was consumed rapidly during the linear phase of growth, this uptake being 90 % complete by day 16 and 100 % complete by day 28. According to Stepan-Sarkissian [16], plant cells synthesize cell wall material and starch from available carbohydrate during the linear growth phase. Moreover, as expected, under the conditions of our work medium conductivity revealed a negative correlation with cell growth during the linear growth phase and the onset of the stationary growth phase (Fig.…”

“…Meristem culture can even improve the technique by eliminating systemic pathogens present in the donor plant (Stepan-Sarkissian, 1990). Therefore, the objective of this study was to develop a micropropagation protocol for P. edulis using meristem as explant.…”
