Selective Transcriptional Down-Regulation of Human Rhinovirus-Induced Production of CXCL10 from Airway Epithelial Cells via the MEK1 Pathway

Abstract: Human rhinovirus (HRV) infections can trigger exacerbations of lower airway diseases. Infection of airway epithelial cells induces production of a number of proinflammatory chemokines that may exacerbate airway inflammation, including CXCL10, a chemoattractant for type 1 lymphocytes and NK cells. Primary human bronchial epithelial cells and the BEAS-2B human bronchial epithelial cell line were used to examine the role of MAPK pathways in HRV-16-induced production of CXCL10. Surprisingly, PD98059 and U0126, two… Show more

“…In addition, we have shown chronic activation of this MAPK pathway upon HRV infection (12,33). As reported earlier, Fra-1 expression is induced upon HRV infection of BEAS-2B cells but is constitutively present in primary HBE cells.…”

“…Thus, we next sought to determine whether the MEK/ERK MAPK pathway was playing a role in Fra-1 expression and consequently in HRV-induced MMP-9 expression. The pharmacological inhibitors of MEK1/2, PD98059 (1-30 mM) and U0126 (0.3-10 mM), which have been shown to inhibit HRV-induced ERK phosphorylation (33), both decreased HRV-induced Fra-1 expression in BEAS-2B cells in a concentration-dependent manner (Fig. 3A, 3B).…”

“…This is particularly interesting in view of a recent publication showing that activation of the MAP kinase kinase (MEK)-ERK pathway by HRV-16 actually inhibits CXCL10 production (30). Thus, after exposure to cigarette smoke, HRV-16 can still effectively induce the MEK-ERK pathway, exerting an inhibitory effect on the expression of CXCL10.…”

“…Activation of the mitogen-activated protein (MAP) kinase kinases SAPK/JNK and ERK was shown to be involved in the regulation of virally induced gene expression (29)(30)(31). SAPK/JNK is involved in the regulation of b-defensin and activates activating transcription factor 2 (ATF2), a component of the Type I IFN response, whereas ERK is involved in the regulation of RV-induced CXCL10 and respiratory syncytial virus (RSV)-induced CCL5.…”
