Chlorpyrifos (CPF) has caused many potential toxicities in nontarget organisms. Fewer studies have been conducted on the effects of lactic acid bacteria (LAB) in mitigating tissue damage induced by CPF in vivo. Therefore, we investigated CPF renal and testicular toxicity and the alleviate effect of probiotic lactobacilli, based on antioxidant and anti‐inflammatory activity, on induced toxicity in an animal model. Biochemical assays showed that CPF induced oxidative stress along with a change in superoxide dismutase (SOD) and catalase (CAT) activity in a tissue dependent manner. After treatment with CPF, testicular and renal levels of TNF‐α were significantly reduced and enhanced, respectively, compared to the control group. Probiotic treatment restored renal and testicular TNF‐α levels and modulated and blocked the increasing effect of CPF on renal IL‐1β levels. Testicular IL‐1β levels in the probiotic‐treated and CPF groups demonstrated similar values. Exposure to CPF significantly induced renal histopathological damage that, of course, was completely inhibited by treatment with Lactobacillus casei and the LAB mixture. In summary, CPF showed significant toxicological effects on oxidative stress and the inflammation rate in CPF‐exposed rats. Therefore, supplementation with probiotic bacteria may alleviate CPF renal toxicity and mitigate its oxidative stress and inflammation effects.

Earlier studies have shown that selenomethionine (SM) supplements in broiler breeders had higher deposition in eggs, further reduced the mortality of chicken embryos, and exerted a stronger antioxidant ability in offspring than sodium selenite (SS). Since previous studies also confirmed that Se deposition in eggs was positively correlated with maternal supplementation, this study aimed to directly investigate the antioxidant activities and underlying mechanisms of SS and SM on the chicken hepatocellular carcinoma cell line (LMH). The cytotoxicity results showed that the safe concentration of SM was up to 1000 ng/mL, while SS was 100 ng/mL. In Se treatments, both SS and SM significantly elevated mRNA stability and the protein synthesis rate of glutathione peroxidase (GPx) and thioredoxin reductase (TrxR), two Se-containing antioxidant enzymes. Furthermore, SM exerted protective effects in the H2O2-induced oxidant stress model by reducing free radicals (including ROS, MDA, and NO) and elevating the activities of antioxidative enzymes, which performed better than SS. Furthermore, the results showed that cotreatment with SM significantly induced apoptosis induced by H2O2 on elevating the content of Bcl-2 and decreasing caspase-3. Moreover, investigations of the mechanism revealed that SM might exert antioxidant effects on H2O2-induced LMHs by activating the Nrf2 pathway and enhancing the activities of major antioxidant selenoenzymes downstream. These findings provide evidence for the effectiveness of SM on ameliorating H2O2-induced oxidative impairment and suggest SM has the potential to be used in the prevention or adjuvant treatment of oxidative-related impairment in poultry feeds.

Wistar rats were divided into 6 numerically equal groups and treated as --Group A: Control; Group B: dimethoate (20 mg/kg b wt); Group C: dimethoate (20 mg/kg b wt) and selenium (0.5 mg/kg b wt); Group D: dimethoate (20 mg/kg b wt) and amla fruit pulp extract (200 mg/kg b wt); Group E: selenium (0.5 mg/kg b wt) and Group F: amla fruit pulp extract (200 mg/kg b wt). Blood samples were collected on 7 and 14 day and serum calcium and phosphate levels were analyzed. Serum calcium levels of rats treated for 7 day with dimethoate (Group B); dimethoate +selenium (Group C) and dimethoate+amla fruit pulp extract (Group D) decreased as compared to control rats (Group A). The administration of amla fruit pulp extract and selenium did not fully restore the reduced calcium levels caused by dimethoate, despite a slight increase in calcium levels of group C and group D. After 14 day, calcium levels decreased after dimethoate; dimethoate+ selenium; and dimethoate + amla fruit pulp extract treatment. Although serum calcium levels are increased in group C and group D as compared to dimethoate treated group (group B) but the levels (group C and D) are still hypocalcemic as compared to control. Levels in group E (selenium) and F (amla fruit pulp extract) remain unaltered on day 7 and 14. Serum phosphate levels decreased after 7 and 14 day in dimethoate, dimethoate+selenium and dimethoate+amla fruit pulp extract treated rats. In group C (as compared with E) and group F (as compared with F) phosphate level decreased. Phosphate levels of group C and group D increased as compared to group B. This indicates that treatment of amla fruit pulp extract and selenium caused phosphate levels to recover.
