Seleno-detergent MAD phasing of leukotriene C4 synthase in complex with dodecyl-β-D-selenomaltoside

Abstract: PDB Reference: LTC 4 S with SeDDM, 3b29.Dodecyl--d-selenomaltoside (SeDDM) is a seleno-detergent with a -glycosidic seleno-ether in place of the ether moiety in dodecyl--d-maltoside. Selenodetergents are candidates for heavy-atom agents in experimental phasing of membrane proteins in protein crystallography. Crystals of a nuclear membraneembedded enzyme, leukotriene C 4 synthase (LTC 4 S), in complex with SeDDM were prepared and a multiwavelength anomalous diffraction (MAD) experiment was performed. The SeDDM … Show more

“…More recently, the human galectin-9 N -terminal carbohydrate recognition domain was crystallized with the aid of selenosugars [ 64 ]. In the same context, dodecyl-β-selenomaltoside has been successfully utilized as a detergent for stabilizing membrane proteins in water [ 65 ].…”

“…In previous SRX experiments, MAD phasing with an Se-labeled detergent was reported with a membrane protein, leukotriene C4 synthase, in complex with dodecyl-β-D-selenomaltoside (35). In another study, although phasing was not successful, binding sites of Br-or Hg-labeled phospholipids or detergents were identified at membrane protein reaction centers (36,37).…”

“…Detergents carrying a heavy atom can be used in membrane protein studies. Dodecyl-β-D-selenomaltoside (SeDDM), a heavy atom analog of dodecyl-β-D-maltoside (DDM), was used to achieve MAD phasing when solving the structure of leukotriene C4 synthase by X-ray crystallography (Figure 5A) [51]. Furthermore, SeDDM was used successfully as a phasing detergent to solve the structure of the prokaryotic pentameric ligand-gated ion channel [52]; however, the sugar headgroups were poorly resolved (Figure 5B).…”
