Self‐Assembly of Amphiphilic Drug–Dye Conjugates into Nanoparticles for Imaging and Chemotherapy

Abstract: An amphiphilic drug-dye conjugate (PTX-Pt-BDP) was designed and synthesized with a platinum compound as the hydrophilic head. The precursor of PTX-Pt-BDP was obtained under mild conditions by means of a three-component Passerini reaction. PTX-Pt-BDP could self-assemble into nanoparticles (PTX-Pt-BDP NPs) in aqueous solution via a nanoprecipitation method. The obtained nanoparticles exhibited favorable structural stability in both water and physiological environment. PTX-Pt-BDP NPs could be endocytosed by cance… Show more

“…Self-assembly of organic molecules to form nanoparticles is more conducive to increasing drug loading and obtaining better water solubility than using drug carriers. [41][42][43] Wang et al [44] combined 18β-glycyrrhetinic acid (GA, 3β-hydroxyl-11oxo-olean-12-ene-29-oic acid) with porphyrins to obtain amphiphilic porphyrins (TPP-GA) (Figure 2). TPP-GA can self-assemble into nanoparticles (TPP-GA NPs) with good water stability in aqueous solutions.…”

“…Multimodal bioimaging can accomplish accurate and rapid diagnosis for tumor nanotheranostic platforms. However, for some tumor treatment modalities such as photodynamic therapy (PDT), [6][7][8][9] sonodynamic therapy (SDT), 10,11 radiotherapy (RT), 12,13 and chemotherapy, [14][15][16] the treatment effect is primarily dependent on the oxygen concentration level. Due to the demand of high oxygen consumption of cancer cells and the lack of oxygen supply caused by abnormal tumor angiogenesis, cancer cells have a hypoxic microenvironment, which is a typical feature of most solid tumors.…”
