To achieve targeted clearance of bacteria and their biofilms, a straightforward strategy to integrate antimicrobial alternating polymers with photothermal polydopamine nanoparticles (PDA NPs) is proposed. By manipulating the alternating distribution of electrostatic, hydrophobic, and hydrogen bonding units in the polymer backbone, the thermal stability of polymer‐particle interaction can be tuned, enabling photothermal‐mediated in situ delivery of cationic antimicrobial polymers into bacteria. The alternating polymer coating significantly enhances the penetration capability of NPs into the Methicillin‐resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli) biofilm, achieving a 99% bactericidal rate within the biofilm at a low concentration after 10 min of near‐infrared irradiation, whereas pristine PDA NPs shows negligible effects. The in‐vitro co‐culture model demonstrates that alternating‐polymer‐loaded NPs selectively eradicate 1 × 107 CFU mL−1 of MRSA and E. coli while preserving over 75% viability of the mammalian cells, including mice fibroblasts, human kidney cells, human cervical cancer cells, and macrophages. The efficacy of these biocompatible NPs in targeting bacteria is further validated in a mouse MRSA‐infected wound model. This approach represents a significant advancement in developing safe and efficient antimicrobial therapies with targeted bacterial killing and minimal off‐target effects based on alternating cationic polymers.

In wound healing and clinical anti‐infection therapy, the current feasibility of nanocatalysts is extremely limited because of inadequate reactive oxygen species (ROS) generation. Herein, a novel H2O2 self‐supplying nanocomposite (M/C/AEK) consists of molybdenum disulfide (MoS2) decorated with calcium peroxide (CaO2) prepared at ambient temperature and encapsulated in AEK hydrogel. In the presence of H2O2 and poly(vinyl pyrrolidone) (PVP), CaO2 nanoclusters, ≈30 nm, are anchored on the MoS2 surface. MoS2/CaO2 can induce both a cascaded peroxidase (POD)‐like and catalase (CAT)‐like catalytic activity to produce toxic hydroxyl radicals through self‐supplied H2O2 and O2 responsive to the faintly acidic environment of acute wounds. The POD‐like activity is increased under acidic compared with neutral conditions, allowing selective treatment of acute, slightly acidic wounds while avoiding the side effects of high‐concentration antibacterial agents on normal tissues. The high near‐infrared photothermal effect synergistically with POD‐like/CAT‐like activity of MoS2/CaO2 boosts the production of more ROS to eradicate Staphylococcus aureus and Escherichia coli bacteria (98.6% and 98.9%) effectively and selectively stimulate wound healing. The porous M/C/AEK hydrogel in the wound microenvironment can efficiently capture bacteria, and its Ca2+ ions and keratin stimulate healing, revealing excellent potential in advanced wound care and infection control therapies.

Diabetic wounds, particularly diabetic foot ulcers (DFUs), are significant threats to human well‐being due to their impaired healing from poor circulation and high blood sugar, increased risk of infection and potential for severe complications like amputation, all compounded by peripheral neuropathy and chronic inflammation. Most therapies and dressings for DFUs focus on one symptom at a time, however, multifunctional smart self‐healing hydrogels can withstand multifactorial motional diabetic wounds. Motional wounds are easy‐to‐split wounds that experience tension, compression, and movement caused by stress now and then. Hyaluronic acid (HA) based self‐healing hydrogels stand out among other biomaterials due to their ability to cover irregular wound surfaces, maintain a moist environment, repair themselves when ruptured, and exhibit excellent biocompatibility. These self‐healing hydrogels can repair damages caused by movement and recover the functional properties during healing. These hydrogels can also act as therapeutic delivery vehicles and tissue regeneration systems. This review demonstrates the potential of HA‐based self‐healing hydrogels for diabetic wound healing. Due to its self‐healing capabilities, these hydrogels offer a customized therapeutic approach for motional diabetic wounds. The review also critically examines the challenges and future directions for HA‐based self‐healing hydrogels in diabetic wound healing.
