Semaphorin 4A as novel regulator and promising therapeutic target in rheumatoid arthritis

Abstract: Rheumatoid arthritis (RA) is a systemic autoimmune disease manifesting in joint destruction. The recognized hallmark of RA pathogenesis is the involvement of immune cells which produce many mediators potentiating an inflammatory environment. RA synovial fibroblasts (RASFs) contribute significantly to disease progression by initiating and regulating many pathways of joint destruction. Detailed molecular insights into RASF biology may lead to identification of important therapeutic targets. The discovery of comm… Show more

“…However, later we and others demonstrated Plexin B1 and/or D1 expression in the immune system, particularly on DC [10,20], T cells [21] and Treg cells (our unpublished observations). As Sema4A regulates the immune response to different antigens such as allergens [19,22,23], infectious agents [14,21,24], and tissue-derived factors in autoimmunity [15,25,26,27,28], it is feasible to conclude that it plays a significant role in anti-tumor immunity that has not yet been assessed.…”

“…Patients with highly active RA (DAS28 > 5.1) and patients with low activity (DAS28 score < 3.2) showed elevated class 4 semaphorin levels. SEMA4A might be involved in RA activity [64]. Elevated SEMA4A levels are directly correlated with DAS28 and DAS28-CRP, markers of the swollen joint count [23,28].…”
