Senolytic Cocktail Dasatinib+Quercetin (D+Q) Does Not Enhance the Efficacy of Senescence-Inducing Chemotherapy in Liver Cancer

Abstract: Hepatocellular carcinoma (HCC) is a leading cause of cancer-related death, which develops in the context of fibrosis and cirrhosis caused by chronic inflammation, in turn due to non-alcoholic fatty liver disease (NAFLD), alcohol consumption and/or hepatitis viral infection. An increased number of senescent cells are associated with age-related tissue degeneration during NAFLD-induced HCC, or during chemotherapeutic treatment. Senolytic agents target selectively senescent cells. A combination of the senolytic d… Show more

“…Interestingly, the appearance and enrichment of DXR‐induced senescence markers were more prominent in HepG2 cells compared with Huh‐7 cells. Huh‐7 cells are more aggressive than HepG2 cells, in vitro and in vivo, and the two cell lines have a different genetic make‐up . While HepG2 cells express wild‐type p53, Huh‐7 cells express a dominant‐negative mutant form of p53 (Y220C), which may be involved in resistance to DXR‐induced p53‐mediated cell senescence …”
