Sensitive time-resolved fluoroimmunoassay for quantitative determination of clothianidin in agricultural samples

Abstract: Europium (Eu(3+))-labeled antibody was used as a fluorescent label to develop a highly sensitive time-resolved fluoroimmunoassay (TRFIA) for determination of clothianidin residues in agricultural samples. Toward this goal, the Eu(3+)-labeled polyclonal antibody and goat anti-rabbit antibody were prepared for developing and evaluating direct competitive TRFIA (dc-TRFIA) and indirect competitive TRFIA (ic-TRFIA). Under optimal conditions, the half-maximal inhibition concentration (IC50) and the limit of detectio… Show more

“…In this study, we established an indirect competition TRFIA for OA in shellfish. Goat anti-mouse IgG, i.e., secondary antibody, was labeled with europium for low detection limit and high sensitivity (Esteve-Turrillas et al, 2010;Sesay et al, 2013;Li et al, 2014;Sheng et al, 2020). Compared with labeling the primary antibody, labeling the secondary antibody avoids the interference of the marker to the binding site of the antigen on the first antibody.…”

“…The fluorophore generates fluorescence at a distinct wavelength when excited by light, which can be quantified using a timelapse fluorometer. The target antibody or antigen is marked using the time-resolved fluorescence immunoassay (TR-FIA) technique, which makes use of lanthanide elements with distinctive fluorescence properties and the associated chelates as markers [18,19]. It frequently makes use of antibody that is adhered to the microplate well's bottom.…”

“…B 0 /IC 50 was selected as important criteria to evaluate the performance of the immunoassays (Brun et al, 2005;Korpimaki et al, 2004). In addition, IC 50 and B 0 also were regarded as primary physicochemical factors to estimate the performance of the methods (Li et al, 2014).…”
