Separation and purification of magnoflorine, spinosin, and 6‴‐feruloyspinosin from Ziziphi Spinosae Semen by high‐speed counter‐current chromatography

Abstract: In the study, high-speed counter-current chromatography was used for separation and purification of magnoflorine, spinosin, and 6‴-feruloyspinosin from Ziziphi Spinosae Semen. With n-butanol-ethyl acetate-water (2:3:5, v/v) as the optimum solvent system, about 75 mg of magnoflorine, 110 mg of spinosin, and 40 mg of 6‴-feruloyspinosin were isolated from 0.5 g of crude extract of Z. Spinosae Semen, with the purity of 95.7, 97.2, and 96.4%, respectively. The chemical structures were identified by MS and NMR spect… Show more

“…Jujuboside A can inhibit cancer cell growth through various mechanisms such as cell cycle arrest, proliferation inhibition, stem cell inhibition, and promotion of aging and can contaminate antitoxins [ 25 , 26 ], and our analysis results show that there are many intersections of targets associated with it and insomnia targets. Spinosin exerts neuroprotective effects by inhibiting oxidative damage and has a significant antidepressant effect, and there is a great association between spinosin and insomnia [ 27 , 28 ]. Although the kinetic values of these two components are relatively low, they have very excellent biological activity and are closely related to our study.…”

“…The isolation and refinement of 6‴-feruloyspinosin, spinosin, and magnoflorine from Ziziphi Spinosae Semen were accomplished through a combination of A TBE-1000A HSCCC and HPLC under the Agilent C-18column (4.6 × 250 mm, 5 μm). The isolated constituents were distinguished via the implementation of MS, 1 H-NMR, and 13 C-NMR spectroscopic analyses [64]. In addition, compared to its one-dimensional counterpart, prep-HPCCCbased multidimensional chromatography stands out with its exceptional ability to isolate and purify.…”

“…Also, the polysaccharide (yield was 1.05 ± 0.08%) was extracted from ZSS with ultrasound-assisted (temperature was 52.5 °C, time was 21.2 min, power was 134.9 W) (Lin et al, 2018). In 2021, Xu et al (2021) separated magnoflorine (75 mg), spinosin (110 mg), and 6‴-feruloylspinosin (40 mg) from ZSS (0.5 g) by high-speed countercurrent chromatography, with a purity of 95.7%, 97.2%, and 96.4%, respectively. In addition, the yields of vitamin E and squalene obtained by the optimized ultrasonic-assisted extraction process were 29.58 ± 0.19 mg/100 g and 820.49 ± 3.66 μg/100 g, respectively (Xiao et al, 2018).…”
