Septal Sealing in the Basidiomycete Coriolus versicolor

Abstract: The way in which the dolipore apparatus contains hyphal damage, and the process of septal sealing have been studied in Corwlus versicolor using combined light and electron microscopy. The technique used allows the structure of septa in adjacent damaged and undamaged hyphae to be compared. The results show that septal sealing, following damage, is a two stage process. The first is the instantaneous plugging of the pore channel with electron-dense material. The second, beginning several minutes later, involves t… Show more

“…Filamentous Ascomycota (i.e., Pezizomycotina) seal their septal pores with Woronin bodies that are associated with the septum (24,32,56). In contrast, SPCs seem to fulfill this role in higher Basidiomycota, viz., Agaricomycotina (1,31,38,50). Several main morphotypes of the SPC exist, namely, vesicular-tubular, imperforate, and perforate, and it seems that they have arisen from the ER or that SPCs and ER both share a common ancestor (51).…”

“…SPCs may act as a sieve to discriminate between organelles that may pass through the septal pore (55), they may guide cytoplasmic streams toward the pore (41), and they may function in protoplasmic streaming to protect the dolipore region in such way that protoplasmic streaming is not disturbed by organelles (10). Alternatively, they may be involved in dolipore plugging (1,31,38,50).…”

“…The formation of electron-dense pore plugs during dolipore synthesis reported elsewhere (Moore & Marchant, 1972) was not evident in this work. These structures are known to be formed as a result of hyphal injury (Aylmore et al, 1984) and play a role in confining the incompatible reaction in non-self fusions (see Aylmore & Todd, 1986). In true dikaryons, nuclear replacement and septum formation apparently serves to maintain the bi-nucleate arrangement, restoring nuclear numbers in compartments that have fused (Aylmore & Todd, 1984a).…”

“…Dolipore septa are elaborate septal complexes found in most fungi but their role in fungal physiology is still not very clear (Gow and Gadd, 1995). Doplipore septa form in situations of extreme 11 cell trauma or stress or even during ageing (Alymore et al 1984). The presence of paranthesomes (Fig 6b) supports this phenomenon because they are generally considered to provide a protective or selective function by regulating the cytoplasm immediately adjacent to the septal swellings and pores (Alymore et al 1984;Gow and Gadd, 1995).…”

Under the severe nutrient limitation conditions imposed by extended operation of the membrane gradostat reactor, it seemed plausible that the formation of dolipore septa played a role in structural support, hyphal tip growth and maintaining rigidity of the hyphae.…”
