Sequence analysis of cDNA coding for a major house dust mite allergen, Der p 1. Homology with cysteine proteases.

Abstract: It has long been recognized that the allergy to mites of the genus Dermatophagoides is associated with diseases such as asthma, rhinitis, and atopic dermatitis (1, 2). In this regard, the species D. pteronyssinus and D. farinae predominate and many studies have been performed to identify the allergens they produce. Studies on D. pteronyssinus have, for example, demonstrated that at least six major allergens are of clinical importance (3, 4). One of the major mite allergens, designated Derp l, has been purified… Show more

“…These allergens appear to be structural homologues and have very similar Nterminal amino acid sequences. The Group II allergens (Derp II and Derf II) are proteins (Me, 15 000) and also heterogeneous on IEF, with almost identical N-terminal amino acid sequences (5,11,34).a The gene encQding Derp I has been cloned and expressed in lambda gt 11 and the full amino acid sequence has been deduced from the cDNA (28). The majority of mite allergic individuals produce IgE antibody to both the Group I and Group II allergens.…”

“…A few examples of wellknown allergens characterized in this way are ragweed allergens Amb a 1 [1] and Amb a 2 [2], grass pollen allergens Lol p 1 [3,4] and Lol p 2 [5], house dust mite allergens Der p 1 [6] and Der f 1 [7], peanut allergen Ara h 1 [8], shrimp allergen Met e 1 [9], and latex allergen Hev b 1 [10]. This technology has the potential to be useful in several ways.…”
