Sequence analysis of plasmid pRA2 from Pseudomonas alcaligenes NCIB 9867 (P25X) reveals a novel replication region

Abstract: The replication region of plasmid pRA2 from Pseudomonas alcaligenes NCIB 9867 (strain P25X) was localized within a 5.9-kbp DNA fragment and its sequence was determined. An interesting feature of the sequence is the presence of a 1.3-kbp region containing seven, highly conserved, direct repeats of 72 bp in length. The pRA2 replication region has two open reading frames (ORFs). ORF1 appeared to be essential for replication and had the potential to encode a novel 30-kDa protein with a predicted helix-turn-helix m… Show more

“…Several amino acid residues conserved between KfrA of pRA2 and IncP-1 plasmids are also conserved in the pRSB101 KfrA-like protein (Kwong et al, 1998). Taking together the findings described above, it is very likely that the KfrA-homologue of pRSB101 and related plasmids plays a role in stable plasmid inheritance and/or plasmidregulation.…”

“…The parABC module of pRIO-5 was identical to that of different IncP6-type plasmids, including pRms149 identified from a clinical Pseudomonas aeruginosa strain (7) and plasmid pRSB105 (57 kb in size) recovered from an active sludge in Germany (18). This module was also 98.7% identical with that of plasmid pRA2 identified in Pseudomonas alcaligenes (10). The repA gene of pRIO-5 encoded a protein identical to that of plasmid pRSB105 and 82% identical to that of plasmid pRms149, which differed in the C terminus.…”

“…A transposase (Noc_A0021), phage integrase (Noc_A0015), and a small number of other phagerelated genes along with restriction modification systems are also found on this replicon. A possible replication protein (Noc_A0039) whose only putative homologue (56% similar over 80% of its length) is the RepA protein encoded by plasmid pRA2 from Pseudomonas alcaligenes (44), together with a possible partitioning system (Noc_A0013 to Noc_A0014), may help this plasmid be maintained within the N. oceani population.…”
