Ser/Thr phosphorylation of hematopoietic specific protein 1 (HS1). Implication of protein kinase CK2

Abstract: Hematopoietic lineage cell-specific protein 1 (HS1), a tyrosine multiphosphorylated protein implicated in receptor-mediated apoptosis and proliferative responses, is shown here to become Ser/ Thr phosphorylated upon incubation of platelets with radiolabeled inorganic phosphate. The in vivo Ser/ Thr phosphorylation of HS1 is enhanced by okadaic acid and reduced by specific inhibitors of casein kinase (CK)2. In vitro, HS1 is an excellent substrate for either CK2 a subunit alone (K m 47 nm) or CK2 holoenzyme, tes… Show more

“…Indeed, LY-294,002 treatment significantly decreased cell viability in first-trimester trophoblast cells at concentrations similar to those observed in previous studies [15,36]. Interestingly, LY-294,002 also inhibits casein kinase 2 [37], a protein kinase that has been shown to protect other cells from apoptosis [38,39]. Because casein kinase 2 was demonstrated previously to phosphorylate and upregulate AKT [40], the results obtained from LY-294,002 treatment confirm a role for the PIK3/AKT pathway in the regulation of trophoblast apoptosis.…”

“…Tyrosine kinases SYK, LYN and LCK and the adaptor protein GRB2 phosphorylate and activate the protein (73)(74)(75)(76)(77)(78). In platelets casein kinase 2 catalyzes HCLS1 threonine and serine phosphorylation (79). A comparison with the phospho-proteome of LPS-treated murine bone marrow derived macrophages (80) revealed that LPS treatment results in diminished HCLS1 phosphorylation, and it might be assumed that regulated phosphorylation causes differential interaction with individual mRNAs (27).…”
