Serological and molecular characterisations of the Egyptian isolate ofBean common mosaic virus

Abstract: Bean common mosaic virus (BCMV) was isolated from the naturally infected bean plants collected from the Kafr El-Sheikh and El-Gharbia Governorates. BCMV induced sever mosaic, vein banding, malformation, leaf curling and stunting on bean plants cv. Giza 6. The isolated virus was propagated in bean plants cv. Giza 6. The identification of BCMV was carried out serologically by an indirect enzyme-linked immunosorbent assay using BCMV antiserum. Positive reaction indicated that the virus under study was related ser… Show more

“…The virions are 750 nm in length with a diameter of 11-13 nm and contain a genomic RNA molecule that is approximately 10 kb long that possesses a 3 0 -terminal polyA tail (El-Sawy et al, 2013;Fang et al, 1995). Virions are principally constructed from CP molecules that are arranged helically around the genomic RNA.…”

“…This has resulted in enhanced interregional seed trade between smallholder farmers and within neighboring countries (Uganda and Tanzania), with minimal phytosanitary regulation (Katungi et al, 2009). In addition, legumes are currently utilised for integrated pest management of whitefly and whitefly-transmitted viruses (Fondong et al, 2000; Lapidot et al, 2014; El-sawy, Mohamed & Elsharkawy, 2014; Uzokwe et al, 2016).…”

“…BCMNV and CABMV are monopartite, single-stranded positive-sense RNA viruses with an average genome size of ∼10 Kb and a polyA tail on the 3′-terminal end (Fang et al, 1995; Mlotshwa et al, 2002; El-sawy, Mohamed & Elsharkawy, 2014; Kehoe et al, 2014a). They encode a single polyprotein that is processed into ten proteins in a similar order and function to all members of the family Potyviridae: P1 (Symptomatology), HcPro (Aphid transmission), P3 (pathogenicity), 6K1, CI (ATPase/RNA helicase, cell to cell movement), 6K2 (Anchoring the viral replication complex to membranes), Nla-Vpg, Nla-Pro Nlb (Genome replication, RNA-dependent RNA polymerase (RdRP) and coat protein (CP) (Aphid transmission, virus assembly, cell to cell and systemic movement).…”
