Background Viral diseases are a matter of great concern for poultry farmers in Pakistan. Multiple common viral respiratory diseases (CVRDs) cause huge economic losses in the poultry industry. The prevalence of CVRDs in many countries, including Pakistan, is not clearly understood. Results Incidences of 5 chicken respiratory viruses: avian influenza virus (AIV), Newcastle disease virus (NDV/AAVV-1), infectious bronchitis virus (IBV), avian metapneumovirus (aMPV) and infectious laryngotracheitis virus (ILTV) were assessed on commercial Pakistani farms with respiratory problems from 2014 through to 2016. While AIV and AAVV-1 were frequently detected (16 to 17% of farms), IBV and aMPV were rarely detected (in 3 to 5% of farms) and ILTV was not detected. We characterized H9 AIV of the G1 lineage, genotype VII AAVV-1, GI-13 IBV, and type B aMPV strains with very little genetic variability in the 2-year study period. Co-infections with AIV and AAVV-1 were common and wild type AAVV-1 was detected despite the use of vaccines. Control measures to limit the virus burden in chicken flocks are discussed. Conclusions Our data shows that AIV (H9), AAVV-1, IBV and aMPV are prevalent in commercial poultry in Pakistan. Further studies are necessary to assess circulating strains, economic losses caused by infections and coinfections of these pathogens, and the costs and benefits of countermeasures. Furthermore, veterinarians and farmers should be informed of the pathogens circulating in the field and hence advised on the use of vaccines.

Background: Avian metapneumovirus (aMPV) is an important viral agent in chicken and turkey production industry and is most commonly associated with acute upper respiratory tract infection and swollen head syndrome. The disease is of significant economic concern as it is highly contagious and can lead to production losses in chicken and turkey flocks, especially when associated with secondary bacterial pathogens. Methods: Herein, we have performed a survey on layer farms in selected areas of Madhya Pradesh State, India to ascertain the aMPV status in laying birds, choanal cleft swabs and sera samples were collected for ELISA and RT-PCR assay. A total of 263 sera and 169 choanal cleft swabs from five districts of Madhya Pradesh State (Indore, Bhopal, Sagar, Guna and Bhind) from layer birds of age group 20-72 weeks were screened for aMPV antibodies and for genome. Result: Out of these Indore had maximum sero-postivity (73.33%), followed by Sagar (5.00%) and Bhopal (4.08%), respectively. Other two districts did not show any seropostivity against aMPV. RT-PCR assay using published primers and in vitro transcribed RNA of nucleoprotein (N) gene as positive control was carried out in 169 choanal cleft samples from four districts (Indore, Bhopal, Sagar and Guna). All the samples were found negative for aMPV genome by RT-PCR assay. Overall, 26/263 (9.88%) sera were positive to aMPV antibodies. As the chickens in India are not vaccinated against avian metapneumovirus therefore, this study indicates that layers are exposed to this important poultry pathogen. This study warrants further investigation in wider geographical area and isolation of aMPV to design the control strategies for aMPV.

Background and Aim: Avian metapneumovirus (aMPV) is a recently discovered respiratory virus in chickens. Avian metapneumovirus has been linked to respiratory syndromes, reproductive failure in affected chickens and turkeys, swollen head syndrome in chickens, and rhinotracheitis in turkeys. Wild birds are considered potential reservoirs of aMPV, particularly aMPV-C. However, little is known about the prevalence of aMPV in Saudi Arabia. Considering the relevance of backyard chickens in the transmission and sustainability of certain avian viral diseases, this study aimed to assess aMPV exposure in backyard chickens and wild birds circulating near selected locations. Materials and Methods: We collected 368 serum samples from unvaccinated backyard chickens in ten locations in Eastern Saudi Arabia. Furthermore, we collected 78 serum samples from species of free-ranging birds belonging to the Columbidae family, such as pigeons and doves, captured from the same areas. Using commercial enzyme-linked immunosorbent assay kits, we tested the sera of domestic backyard chickens and wild birds for antibodies against aMPV. Results: Our results showed that 74/368 birds were positive for aMPV-related antibodies. Conversely, none of the tested wild birds seroconverted to aMPV. Conclusion: The antibody titers detected in the backyard chickens suggested recent exposure to aMPV. Considering these results, further large-scale serological and molecular studies are needed to evaluate the prevalence of aMPV in these birds and characterize the circulating strains of aMPV in this region.
