Seroprevalence of neutralizing antibodies against adenovirus type 14 and 55 in healthy adults in Southern China

Abstract: Re-emerging human adenovirus types 14 (Ad14) and 55 (Ad55) have caused severe respiratory diseases and even deaths during recent outbreaks. However, the seroprevalence of neutralizing antibodies (nAbs) in healthy adults, which may reflect previous circulation and help to predict potential outbreaks, remains unclear. In this study, we established micro-neutralizing (MN) assays on the basis of recombinant Ad14 and Ad55 reporter viruses, and we investigated serum nAbs in healthy blood donors from Southern China. … Show more

“…Similar distributions in seropositive rates and nAb titers of HAdV-4 or HAdV-5 between gender groups were observed (Fig. 3 ), in accordance with the results observed for other adenovirus types 4 , 29 . These results suggest that the sensitivity of men and women to infection by HAdV-4 or HAdV-5 was consistent and that the same level of nAbs could be generated by males and females after infection with HAdV-4 or HAdV-5.…”

“…To date, a total of 90 HAdV subtypes have been documented and classified into 7 species (A–G) according to their serological and molecular characteristics 1 3 . A variety of severe diseases with a broad range of symptoms, such as acute febrile respiratory disease, conjunctivitis, cystitis, gastroenteritis, myocarditis, and even life-threatening pneumonia are caused by HAdV infections in healthy adults and particularly in immunocompromised infants or young children 2 , 4 .…”

“…The titers of neutralizing antibodies against Ad5 and Ad2 were detected with an MN assay as previously described 46 . Briefly, HEK293 cells were seeded into 96-well plates at 3 × 10 4 cells per well.…”

“…Outbreaks of HAdV-associated ARI (HAdV-ARI) in adults have been reported in communal living populations, including school, hospital, and military populations [3][4][5][6][7][8]. HAdV-55, a novel recombinant strain of HAdV-11 and HAdV-14 that causes ARI in adults [9], emerged as an epidemic strain in Asian and European countries from 2004 [6,8,[10][11][12][13][14][15][16][17][18]. In the Republic of Korea, fatal cases of HAdV-55 pneumonia among military personnel have been reported since 2012 [19], and a large military outbreak associated with HAdV-55 was observed in the winter of 2014 [20].…”
