Serpentinimonas gen. nov., Serpentinimonas raichei sp. nov., Serpentinimonas barnesii sp. nov. and Serpentinimonas maccroryi sp. nov., hyperalkaliphilic and facultative autotrophic bacteria isolated from terrestrial serpentinizing springs

Abstract: Three highly alkaliphilic bacterial strains designated as A1T, H1T and B1T were isolated from two highly alkaline springs at The Cedars, a terrestrial serpentinizing site. Cells from all strains were motile, Gram-negative and rod-shaped. Strains A1T, H1T and B1T were mesophilic (optimum, 30 °C), highly alkaliphilic (optimum, pH 11) and facultatively autotrophic. Major cellular fatty acids were saturated and monounsaturated hexadecenoic and octadecanoic acids. The genome size of strains A1T, H1T and B1T was 2 5… Show more

“…In addition, the relative abundances of Hydrogenophaga/Serpentinomonas and unclassified Comamonadaceae on mica schist were the highest in the methanol-amended microcosms. Hydrogenophaga and Serpentinomonas strains have been shown to grow on acetate and formate ( Bird et al, 2021 ). Correspondingly, one mica schist epilithic Serpentinomonas MAG had genes for acetate metabolism ( Nuppunen-Puputti et al, 2022 ).…”

“…While calcium carbonate is mostly insoluble and thus unavailable as a carbon source, it has nevertheless been shown to support the growth of some serpentinite-hosted microorganisms. This might be the result of local redissolution into bicarbonate catalyzed by the carbonic anhydrase ( Suzuki et al, 2014 ; Fones et al, 2019 ; Bird et al, 2021 ) ( Table 1 ). Alternatively, small organic molecules may serve as primary source of carbon.…”

“…Transferred across the cell membrane via specialized transporters, formate can be oxidized to CO 2 via the formate dehydrogenase in the pH-neutral cytoplasm ( Brazelton et al, 2022 ). Likewise, bicarbonate can be reduced to CO 2 via the carbonic anhydrase ( Suzuki et al, 2014 ; Bird et al, 2021 ). The produced CO 2 is subsequently introduced to different carbon fixation pathways yielding acetyl-CoA.…”

“…Afterward, Macromonas bipunctata , another species of the genus Macromonas , was described by Dubinina and Grabovich [ 5 ] with a strain isolated from the precipitates of sewage aeration tanks. Due to the absence of available cultures and 16S rRNA gene sequences for M. mobilis , only Macromonas bipunctata has been designated as the type species of the genus for taxonomic classification [ 6 , 7 ]. The type strain Macromonas bipunctata IAM 14880 T exhibits characteristics such as being Gram-stain-negative, non-pigmented, oxidase-positive, catalase-negative, motile due to polar flagella, and aerobic.…”
