Serum Bile Acids Profiling in Inflammatory Bowel Disease Patients Treated with Anti-TNFs

Abstract: Background: Inflammatory bowel diseases (IBD), ulcerative colitis (UC), and Crohn’s disease (CD), represent systematic chronic conditions with a deficient intestinal absorption. We first attempt to investigate the serum bile acids (sBAs) profile in a large cohort of IBD patients to evaluate changes under anti-TNF alpha treatment. Methods: Forty CD and 40 UC patients were enrolled and BAs were quantified by high-pressure liquid chromatography-electrospray-tandem mass spectrometry (HPLC-ES-MS/MS). Up to 15 diffe… Show more

“…They found significantly increased GCA concentration in the serum of HCC patients. Apart from hepatobiliary diseases, serum BAs were applied to distinguish BA profiles between healthy controls and patients in end‐stage renal disease [89] or inflammatory bowel disease [90,91]. Furthermore, some research groups focused on how serum BA composition is affected by lipid‐rich diet and a peak in total BAs was observed 2 h after intake [79,92].…”

“…Furthermore, 13 C-NMR, following incorporation of a labelled precursor, enables the interrogation of cellular pathways such as glycolysis and the tricarboxylic acid (TCA) cycle. 31 P-NMR allows for the detection of metabolites that provide insight into energy or phospholipid metabolism [61,63]. Unlike MS, identification of metabolites by NMR can be performed without prior separation of compounds in the sample.…”

“…Examples of commonly used multivariate discriminant tests in metabolomic studies are partial least-squares discriminant analysis (PLS-DA) [24,33,34,41,46,52,53] and principal component analysis (PCA) [13,16,31,34,46,52]. PLS-DA, a multivariate dimensionalityreduction tool, is a supervised classification method, in which the samples are designated into their classes for comparison.…”

“…Even a slight difference in BA hydroxylation patterns may exert drastically different biological responses (Li and Chiang 2009;Song et al, 2015). Therefore, BA pro le assessments are garnering increasing interest to screen potential biomarkers for the early diagnosis of human liver and intestinal diseases (Fu et al 2020;Slopianka et al 2017), malnutrition (Lin et al 2020), and healing processes (Roda et al 2019). It was noticed that the BA pro les of sh could be in uenced by diet (Zhang et al 2017), however, it was unclear about the agent in diet that exerted the effect.…”
