Serum concentrations of flurbiprofen in rheumatoid patients receiving flurbiprofen over long periods of time

Abstract: Serum concentrations of flurbiprofen were measured following single oral doses of 50 mg to 200 mg in patients who previously had received flurbiprofen over long periods of time. The apparent elimination half-lives in groups of patients who received different doses were not significantly different from each other nor from the value previously obtained in a group of healthy volunteers given a single oral dose of 50 mg. Areas under serum concentration versus time curves were linearly related to dose over the rang… Show more

“…In the present study, the mean plasma concentration was 3331 ng ml -1 in the patients receiving orally administered flurbiprofen. This result is comparable with the previous data [9,[11][12][13][14]. Concerning topical application with the same two tapes as used in the present study, it was reported that the Cmax in the plasma was 870 ng ml -1 at the tmax of 11.6 h [8].These data are also comparable with the present result (1369 ng ml -1 ) on the plasma concentration in the patients receiving topically applied flurbiprofen.…”

“…After oral administration of a tablet containing 40 mg flurbiprofen, the Cmax in the plasma was 5600 ng ml -1 and the tmax was 1.4 h [9]. After oral administration of a tablet containing 50 mg flurbiprofen, several studies reported that the Cmax ranged from 4200 to 8200 ng ml -1 and that the tmax ranged from 1.3 to 2.8 h [11][12][13][14]. In the present study, the mean plasma concentration was 3331 ng ml -1 in the patients receiving orally administered flurbiprofen.…”

“…The elimination half-life of flurbiprofen is 3 to 4 hours and does not seem to be significantly different following long term administration (Adams et aI., 1973;Cardoe et al, 1977).…”

“…Intraday preci- Table 3 Regression equations, limits of detection (LOD), and limits of quantitation (LOQ) for six NSAIDs in human plasma using the present method a Data given as mean ± SD calibration are summarized in Table 6. These concentrations in plasma were within therapeutic levels [22,[30][31][32][33][34].…”

“…The LOD values of all drugs under optimal conditions were 0.002-0.005 μg/ml; the LOQ values were 0.01-0.02 μg/ml. The therapeutic levels of NSAIDs were reported to be 16.1 μg/ml for alminoprofen [30], 11.3 μg/ml for zaltoprofen [22], 15.3-53 μg/ml for tiaprofenic acid [31], 4.45 μg/ml for pranoprofen [32], 17-49 μg/ml for ibuprofen [33], and 8.2-14.6 μg/ml for fl urbiprofen [34]. Therefore, our present method is sensitive enough to analyze their therapeutic levels.…”
